摘 要:目的:研究RIZ1基因mRNA及蛋白在人食管鳞癌中的表达。方法:以48例人食管鳞癌组织、相应癌旁及正常组织为研究对象,Real-time PCR法检测RIZ1基因mRNA表达情况;55例癌组织及相应正常组织采用免疫组织化学法检测RIZ1蛋白的表达情况。结果:统计结果显示人食管鳞癌组织中RIZ1 mRNA的表达量显著低于正常食管组织(P<0.01),癌旁组织中的表达量也显著低于正常食管组织(P<0.01),但癌组织与癌旁组织的表达量无显著性差异(P=0.067)。RIZ1蛋白在癌组织中阳性表达率为29.1%(16/55),在正常食管组织中阳性表达率为76.4%(42/55),差异具有统计学意义(P<0.01)。结论:人食管鳞癌组织中RIZ1基因不仅在mRNA转录水平,而且在蛋白表达水平明显减低。提示RIZ1表达降低可能与食管鳞癌的发生有关,RIZ1在食管鳞癌的发生过程中起到抑制作用。RIZI可能是食管鳞癌的潜在抑制基因。Abstract Objective: To observe the expressions of retinoblastoma protein- interacting zinc finger gene 1 ( RIZ1 ) mRNA and protein in human esophageal tissues. Methods: The human esophageal tissues including the cancer tissues, which correspond to neoplastic tissues, and normal esophageal tissues were obtained from 48 patients with esophageal squamous cell carcinoma ( ESCC ). The RIZ1 mRNA expression levels were investigated using real-time polymerase chain reaction. The tissues from 55 cases were surgically obtained, including the cancer tissues and corresponding normal esophageal tissues. Specific RIZ1 antibody staining was performed to observe the RIZ1 protein expression. Results: Compared with the normal tissues, the RIZ1 mRNA expression level was significantly lower in the cancer tissues ( P 〈0.01 ) and in the adjacent tissues ( P 〈0.01 ). However, no significant differences between the cancer and normal adjacent tissues were observed ( P = 0.067 ). The RIZ1 protein expression level observed in the cancer tissues was 29.1 % ( 16 / 55 ),whereas the value was 76.4 % ( 42 / 55 ) in the normal tissues ( P 〈 0.01 ). The two groups were statistically different. Conclusion: The RIZ1 gene expression level is not only lower in transcription but also in the ESCC protein. RIZ1 expression reduces ESCC occurrence, which indicates that RIZ1 is a candidate tumor suppressor gene.
