摘  要:目的:研究并比较DREAM蛋白的2个主要亚型DREAM A和DREAM B在小鼠多种组织尤其是神经组织的表达水平的差异。方法:取不同年龄小鼠的各种组织匀浆后提取RNA,采用real time PCR的方法检测各种组织中DREAM蛋白2个亚型的mRNA的表达水平。结果:DREAM的两个亚型A和B在神经和非神经组织中广泛存在。在大脑皮层、小脑、海马及下丘等神经组织中DREAM A的表达水平要远高于DREAM B,并且在小鼠上述神经组织中DREAM A随着动物发育成熟,表达水平会逐渐升高,而DREAM B在神经组织发育初期呈较高水平表达。结论:DREAM A是成年小鼠中DREAM的主要亚型,而DREAM B可能在动物的发育早期发挥一定作用。Objective: To study and compare the expressional difference of DREAM's isoforms A and B in different tissues, especially in nerve tissues of mice. Methods: RNA of some non-nerve tissues and some nerve tissues were extracted, then detected the mRNA level in non-nerve tissues and nerve tissues of mice by real time PCR. Results: Both DREAM A and B were widely distributed in nerve tissues and non-nerve tissues. The expressional level of DREAM A was extremely higher than DREAM B in nerve tissues such as cortex, cerebellum, hippocampus and hypothalmus. It is also found that the level of DREAM A was gradually increase with the development of the mice in nerve tissues, but DREAM B showed the highest level in immature stage. Conclusion: DREAM A is the major isoform in mature mice, but DREAM B may play a role in the early stage of development.
