摘 要:目的:探讨瘦素对角质形成细胞增殖的作用及其创面愈合中的可能作用及机制。方法:原代培养人表皮角质形成细胞并鉴定。分别观察瘦素对角质形成细胞的时间梯度作用、浓度梯度作用、JAK抑制剂AG490对瘦素促增殖作用的影响,培养24h后收集细胞行MTT检测、CK17及CK19免疫细胞化学染色。结果:24、48、72h后,瘦素作用组均较对照组有明显的细胞增殖。在0~400ng/mL浓度范围,瘦素干预组细胞均较对照组MTT检测OD值升高,有明显的细胞增殖;但在800ng/mL浓度,瘦素作用组较对照组细胞数更少。CK17及CK19免疫细胞化学染色,瘦素组较对照组着色更深、着色细胞数更多;而AG490+瘦素组较瘦素组着色更浅、着色细胞数更少。结论:100ng/mL瘦素对角质形成细胞有时间依赖性促增殖作用。0~400ng/mL浓度范围内,瘦素对角质形成细胞有浓度依赖性促增殖作用。AG490能有效抑制瘦素对角质形成细胞的促增殖作用,提示JAK2/STAT3信号途径参与瘦素的跨膜信号传导。Objectives The aim of this study was to elucidate the effect of leptin on keratinocytes proliferation, the possible role and the mechanism of leptin on wound healing. Methods Primary cultured and identificated human keratinocytes by CK17 and CK19 immunocytochemistry staining. To observe the dose- and timedependent role of leptin on keratinocytes, and the impact of JAK inhibitor (AC,490) on leptin induced keratinocytes proliferation, cells in all groups were collected and examined by MTr assay after 24 h incubation with leptin. Results After 24, 48 and 72 h incubation in D-KSFM with 100 ng/mL leptin, cell proliferation in the leptin intervention group was higher than that in the control group. When the therapeutic concentration range of leptin was 0-400 ng/mL, OD value of cells in leptin group is higher than that in control group. When the concentration of leptin was 800 ng/mL, however, there were fewer cells in the leptin intervention group compared with the control group. The results of immunocytochemistry staining showed that the positive expression of CK17 in leptin group was stronger than in the control group and AG490 + leptin group. Conclusions The positive effect of leptin on keratinocytes proliferation showed a tendency of time-dependent. Meanwhile, there was a concentration-dependent proliferation of leptin on keratinocytes when concentration range is from zero to 400 ng/mL. AG490 can effectively inhibit leptin induced keratinoeytes proliferation, which suggested that JAK2/STAT3 signal pathways involved in transmembrane signal transduction of leptin.
