摘  要:目的探讨磁共振背景信号抑制弥散加权成像(MR DWIBS)在评价磁标记负载肝癌抗原的树突状细胞疫苗活体内移植后对原发性肝癌的治疗作用。方法用粒/巨细胞集落刺激因子(GM-CSF)和白细胞介素4(IL-4)诱导分化DC获得DC疫苗。并且用超顺磁性氧化铁标记.建立兔VX2肝癌模型,皮下注射磁标记的DC疫苗,观察DC疫苗的体内分布。运用DWIBS不同时间点活体检测肿瘤的大小、形态。结果①DC疫苗的体内分布:注射树突状细胞疫苗后,兔肝细胞内可见较多蓝染铁颗粒沉着,而仅有少数Kupffer细胞内可见蓝染铁颗粒沉着;②肿瘤的DWIBS影像上,肝实质信号减低最显著,肿瘤边界清楚;③同时间点治疗组与对照组的凋亡指数(AI)情况:治疗后1周、2周治疗组分别与相应对照组凋亡指数差异有统计学意义(P<0.05);④不同时间点治疗组及对照组的ADC比值情况:治疗后1周、2周治疗组分别与相应对照组ADC比值差异有统计学意义(P<0.05)。结论磁标记负载肝癌抗原的Dc疫苗体内能诱导产生明显的抑瘤作用。而用DWIBS成像技术则为原发性肝癌的免疫基因治疗提供了重要的形态学信息。Objective To investigate the MRI DWIBS imaging in the evaluation of effectivenesse of magnetic labeled den- dritic cell vaccine in treatment of rabbit liver VX2 tumor. Methods In this work, GM-CSF and IL-4 were employed to in- duce the differentiation of dendritic cell (DC) to gain DV vaccine, which was further labeled with superparamagnetic iron oxides. The rabbit VX2 liver cancer model was then established and the magnetic labeled DC vaccine was injected into this model. The electrical microscopy was used to prove DC vaccine distribution in vivo. The changes in tumor volume and shape were observed by MRI DWIBS at different time points. Results (1) Distribution of DC vaccine in vivo., after the ad- ministration of IX; vaccine, many blue iron deposits were seen in hepatocyte of liver tissue, whereas, only appeared in few kupffer eells; (2) MRI imaging of the liver tumor., there was a marked decrease in liver signal intensity on all post-contrast T2-weighted images. The tumor-liver contrast on GRET2WI was best; (3) Cell apoptosis for hepatic VX2 tumors after ad- ministration of DV vaccine at different times: on the 7th day, 14th day, the apoptotic index in the therapeutic group and control group were compared, with significant difference, P 〈0. 053 and (5) The ratio of ADC values in hepatic VX2 tumors after administration of DC vaccine at different times., on the 7th day, 14th day, the ratio of ADC values in therapeu- tic group and control group were compared, with significant difference, P 〈0.05. Conclusion DV vaccine by SPIO in hepatocellular carcinoma can inhibit the growth of human HCC in vitro. MRI DWIBS imaging technology may provide im- portant morphological information for immune gene therapy of primary liver cancer.
