摘  要:目的:观察高频重复经颅磁刺激(rTMS)对急性局灶性脑梗死大鼠运动感觉功能及患侧室管膜下区(SVZ)神经干细胞(NSCs)增殖的影响。方法:48只大鼠随机分为假手术组、脑梗死组和rTMS组各16只,采用大脑中动脉阻塞再灌注(tMCAO)法制作急性脑缺血大鼠模型,假手术组不造成缺血,rTMS组给予rTMS治疗。在治疗前和治疗后7、14d时,给予运动感觉功能评估;使用免疫组织荧光化学法检测患侧大脑SVZ BrdU+/Nestin+细胞密度。结果:治疗后7d,rTMS组较脑梗死组运动感觉功能明显改善(P<0.05),14d各组间无明显差异。患侧SVZBrdU+/Nestin+细胞密度在治疗后7d,脑梗死组较假手术组明显增高(P<0.05),rTMS组较脑梗死组及假手术组均显著增高(P<0.001);14d时rTMS组与假手术组比较,差异有统计学意义(P<0.05)。结论:rTMS治疗可促进急性脑缺血大鼠运动感觉功能恢复及NSCs增殖。Objective: To investigate the effect of rTMS with high frequency on recovery of sensor-motor function and expression of neural stem cells in ipsilesional subventricular zone(SVZ) of rats subjected to focal cerebral ischemia.Methods: A rat model of acute focal cerebral ischemia was established using transient middle cerebral artery occlusion(tMCAO) technique.Forty-eight rats were randomly assigned to the sham,ischemia and rTMS groups.rTMS treatments were given to rTMS group per day since 1 d after tMCAO.Sensorimotor function was evaluated before the treatment and on days 7 and 14 after the treatment.The proliferation of NSCs in the ipsilesional SVZ was identified by double immunofluorescence staining(BrdU/Nestin).Results: After 7 days’ treatment,the sensor-motor function and the density of BrdU+/Nestin+ cells of rats in rTMS group had significantly improved compared with those in the sham and ischemia groups.After 14 days,there was significant difference was found between the sham and rTMS groups.Conclusion: rTMS can promote the recovery of sensormotor function and proliferation of neural stem cells after acute focal cerebral ischemia.
