摘 要:目的研究超顺磁性氧化铁(superparamagnetic iron oxides,SPIOs)Feridex及Resovist对神经干细胞(neural stem cells,NSCs)生物学特性的影响。方法 NSCs培养及鉴定;使用Feridex及Resovist制备磁标记NSCs;利用免疫细胞化学、透射电镜和Prussian blue染色等方法对磁标记NSCs的生长、分化等生物学特性进行研究。结果 Feridex及Resovist分别与NSCs共同孵育后,透射电镜及Prussian blue染色显示胞浆中含有铁颗粒,SPIOs也可以随细胞的分裂增殖而传到子代细胞中。随Feridex及Resovist浓度的增高(2.8μg/mL~16.8μg/mL),Feridex及Resovist对NSCs存活、分化能力的影响无显著性差异(P>0.05)。当Feridex及Resovist的浓度大于22.4μg/mL时,SPIOs影响其存活和分化(P<0.05),在同一浓度条件下,Feridex和Resovist对NSCs的影响无显著性差异(P>0.05)。结论低浓度(<22.4μg/mL)的Feridex及Resovist对NSCs生物学特性无明显影响。Objective To study the biological characteristics of Feridex or Resovist labeled neural stem cells (NSCs). Methods The NSCs were cultured and differentiated from newborn rat brain. NSCs were labeled with Feridex or Resovist to be magnetic labeled NSCs. With im- munocytochemistry, transmission electron microscopy (TEM) and Prussian blue dye, the growth, differentiation and other biology characteristics of magnetic labeled NSCs were studied. Results When NSCs were incubated with Feridex or Resovist, the iron particles were seen in intracellular (non-nucleus) and also existed in dauhgter clones. The of NSCs had no obvious difference with lower concentration of Feridex growth and differentiation or Resovist (2.8 μg/mL - 16.8 μ/mL) (P 〉0.05). NSCs had obvious difference with higher concentration of Feridex or Resovist ( ≥22.4 μh/mL) (P 〈0.05 ) ,and had no obvious difference with same concentra- tion of Feridex or Resovist (2.8 μg/mL - 16.8 txg/mL) (P 〉 0. 05 ). Conclusion Lower concentration of Feridex or Resovist ( 〈 22.4 μg/mL) has no obvious effect on the biology characteristics of NSCs.
