摘  要:目的研究胃癌组织中Syk基因启动子甲基化与术后复发转移的关系。方法采用甲基化特异性PCR法检测胃癌组织及癌旁正常胃组织的Syk基因启动子甲基化状态。结果在70例胃癌组织中,Syk基因启动子甲基化发生率为45.7%(32/70);在癌旁正常组织发生率分别为0(0/70);在胃癌组织中,Syk基因甲基化发生率明显高于癌旁正常组织(P<0.001)。检测到Syk基因启动子甲基化的32例病例中,有21例病例术后发生复发转移,发生率为65.6%(21/32);在未检测到Syk基因启动子甲基化的38例病例中,仅有7例病例术后发生复发转移,发生率为18.4%(7/38)(P<0.001)。结论 (1)Syk基因启动子甲基化与胃癌术后复发转移密切相关。(2)胃癌组织中Syk基因启动子甲基化的检测有助于胃癌术后复发转移的早期诊断及预后评估。Objective To research of the relationship between the aberrant methylation of Syk gene and recurrence and metastasis after resection of gastric cancer. Methods By using methylation-specific polymerase chain reaction technique (MSP), methylation of Syk promoter region from gastric cancer patients (tumor tissues, adjacent normal tissues) were detected. Results The frequency of methylation of Syk promoter region was 45.7%(32/70)in gastric cancer.In contrast, the frequencies of methylation of Syk promoter region were 0(0/70)in adjacent normal tissues.The rates of methylation of Syk promoter in gastric cancer was higher than that of adjacent normal tissues (P〈0.001).The frequency of postoperative recurrence and metastasis in 32 cases with Syk promoter methylation was revealed in 34.4% (11/32) and that in 38 cases with Syk promoter unmethylation was revealed in 18.4% (7/38)(P〈0.001).Conclusion (1)Syk promoter methylation may play an important role in postoperative occurrence and progression of gastric cancer.(2)Detecting the expression of Syk gene in gastric carcinoma could improve the early diagnosis of gastric cancer and evaluate the prognosis.
