摘 要:目的探讨甘露醇干预对大鼠急性缺血性脑卒中大脑不同时期不同部位星形胶质细胞活性的影响。方法采用改良型线栓法实施左侧大鼠大脑中动脉阻塞(middle cerebral artery occlusion,MCAO)2h后再灌注建立脑缺血再灌注模型。随机将大鼠分成空白组、假手术组和缺血再灌注模型组,各12只。每组再随机分成甘露醇组(甘露醇最小有效剂量即10mg/kg大鼠尾静脉缓慢注射,每6h 1次)和非甘露醇组(注射等量的无菌注射用水),各6只,分别于手术后24h和72h处死解剖,取脑组织切片做免疫组化染色,计算缺血周边、海马CA1区的骨架蛋白即胶质纤维酸性蛋白(glial fibrilary acidic protein,GFAP)阳性细胞数,通过观察GFAP阳性数目研究缺血性卒中后缺血周边、海马CA1区星形胶质细胞(astrocyte,AS)的增殖情况。结果在大鼠空白组、假手术组和缺血再灌注模型组中,缺血再灌注模型组中的甘露醇组比各组的非甘露醇组缺血周边区域、海马CA1区域GFAP阳性细胞数明显增多,具有显著统计学意义(P<0.01),并随时间的延长而增加(P<0.05)。结论大鼠缺血再灌注组在甘露醇干预24h和72h后,其缺血周边区域和海马CA1区域GFAP阳性细胞数表达增强,AS活性增加,增生活跃,并随时间的延长而增加,提示甘露醇除脱水降颅压外,还参与了大鼠缺血再灌注模型组缺血周边及海马等区域的AS增殖反应。Objective To investigate astrocytes activity effect with Mannitol in rats of acute ischemic stroke during different periods and different parts of the brain. Methods Implementation of the left middle cerebral artery occlusion (MCAO)by reformed suture emboiization and reperfusion after 2h,to make the cerebral ischemia-reperfusion model of rats. According to l:hI rats were randomly divided into the blank control group, sham operation group and ischemia-reperfusion model group of 12 rats each. Each group to random into mannitol group (mannitol least effective dose that 10 mg/kg big rat tail intravenous injection, slow every 6h) and the non-mannitol group (an injection of same amount of sterile water) every 6 rats,The rats were executed and anatomized after 24h and 72h respectively and were taked the brain tissue slice do immunohistochemical stains, calculated, seahorses around the CA1 ischemia scaffolding proteins called ghal fibers acid protein (GFAP) positive cell count, through the observation of the number GFAP positive study acute ischemic stroke, seahorses CA1 area around ischemia astrocytes (AS) proliferation. Results Rat blank control group, sham operation group and ischemia-reperfusion model group,the ischemia reperfusion model group in the mannitol group than groups of non mannitol group ischemia surrounding areas ,the hippocampal CA1 region GFAP positive cells were significantly increased,significantly statistically Significant(P〈0.01),and increased with time(P〈0.05). Conclusion Ischemia-reperfusion group mannitol intervention 24h and 72h after the ischemia peripheral region and the hippocampal CA1 region of GFAP positive cells express enhanced, AS activity increased, hyperplasia, and increased with time ,prompt mannitol lowering the intracranial pressure, is also involved in ischemia reperfusion model group,ischemia peripheral and hippocampal region AS proliferation response.
