小鼠卵巢玻璃化冻存不同时段FSH干预对Cx43表达的影响

摘　　要：目的通过在玻璃化冻存不同阶段给予小鼠卵巢组织促卵泡刺激素(FSH)干预,观察FSH对缝隙连接蛋白Connexin-43(Cx43)表达的影响,从而寻找最佳的FSH干预途径。方法将4周龄C57BL/6J小鼠卵巢组织分为新鲜对照组(CG)、玻璃化冻存对照组(VCG)、0.3 IU/mL FSH干预玻璃化冻存组,包括全程添加FSH组(OG-FSH)、冻存前添加FSH组(FG-FSH)以及冻存后添加FSH组(LG-FSH)。通过形态学、免疫组织化学技术及Western-blot技术观察并分析各组卵巢组织Cx43蛋白表达情况。结果正常卵泡百分比为玻璃化冻存数量最低,全称FSH干预玻璃化冻存组最高(P<0.05),全程添加FSH组(OG-FSH)最高,而其他组间正常卵泡百分比间的比较差异无统计学意义。各组间Cx43蛋白表达量从高到低,依次为全程添加FSH组(OG-FSH)、冻存前添加FSH组(FG-FSH)、冻存后添加FSH组(LG-FSH)、新鲜对照组(CG)、玻璃化冻存对照组(VCG),且差异有统计学意义(P<0.05)。结论玻璃化冻存全程添加FSH更有利于卵巢组织形态结构的保持以及Cx43蛋白的表达。Objective To find out the best way of vitrafieation intervention of FSH via observation the expression of Cx43. Methods 4 -week -old C57BL/6J mouse were divided into three groups： control group （CG）, the ovaries come from normal mice; vitrification control group （VCG）, no FSH in the medium; vitrification group intervention by 0.3IU/mL FSH including the treatment by FSH in the overall process of vitrification （ OG - FSH）, in the front process of vitrification （ FG - FSH） and in the later process of vitri- fication （ LG - FSH）. The expression of Cx43 were observed and analyzed in each group by the morphology, immunohistochemical tech- nique and western - blot technique. Results The percentage of normal follicles in the VCG was the lowest （P 〈0. 05）, it had a signifi- cant statistical difference from other groups. The results of immunohistochemical technique and western - blot technique shown that the expression of Cx43 from high to low in turn was OG - FSH, FG - FSH, LG - FSH, CG and VCG, respectively, there were significant difference from each other （ P 〈0. 05）. Conclusion The overall process of vitrification used FSH may be more conducive to keep the ovary morphological structure and the expression of Cx43.
