摘 要:以吡嗪作为内标物,建立核磁共振氢谱(1H NMR)测定橙皮苷含量的方法。首先通过分析橙皮苷的1H NMR谱图,考察仪器的稳定性以及脉冲宽度、延迟时间、扫描次数等影响测定结果的参数;其次设定峰面积截取方法,选择定量用特征峰,根据特征峰面积积分值计算样品含量。橙皮苷定量分析最佳参数为:室温,匀场3次,P1=10μs,D1=1.5μs,Ns=32。选定吸收峰C8-H峰为定量用特征峰。该方法使峰面积截取值固定,避免因操作人员的不同而造成的主观误差。With pyrazine as an internal standard,determination of hesperidin content using 1H nuclear magnetic resonance was created.First of all,through the analysis of the 1H NMR spectra of hesperidin,the stability of the instrument and parameters including the pulse width,delay time and the number of scans were studied;second,intercept method of peak area was set,the characteristic peaks used for quantification was selected,hesperidin content was calculated based on peak area values.The optimal parameters were as follows:room temperature,shimming 3,P1=10 μs,D1=1.5 μs,Ns=32.C8-H peak was chosen as the characteristic peak.This method fixed peak area values,avoiding operator errors caused by different operators.
