摘  要:目的分析HCT-116人结肠癌裸鼠动物模型体内微量蛋白谱,为结肠癌的早期诊断寻找特有的微量蛋白标志物。方法按随机分配原则,将20只BALB/C-nu,4周龄的裸小鼠分成实验组和对照组各10只,建立HCT.116人结肠癌裸鼠动物模型,在不同时间采集稞鼠血标本,用PBSII/C型蛋白质指纹图谱仪检测标本中的微量蛋白谱,通过CiphergenPro—teinehip3.0和BiomarkerWizard3.1软件自动采集和分析数据,寻找差异表达蛋白。结果在实验组和对照组问发现120个蛋白质,这些蛋白质在实验组和对照组间的差异具有统计学意义(P〈O.01),但具有规律性变化的有5个,其质荷比分别为2357Da,2424Da,2637Da,8183Da和10133Da,其中2637Da,8183Da和10133Da三种微量蛋白在接种后裸鼠体内表达逐渐增强,2357Da和2424Da两种微量蛋白在接种后裸鼠体内表达逐渐减弱,通过蛋白数据库查询,了解到以上蛋白在诱导细胞有丝分裂、增强钙离子活性、参与细胞迁移和转录调控等方面具有重要作用。结论在HCT-116人结肠癌裸鼠动物模型体内具有特异的微量蛋白表达,所发现的5种具有规律性变化的微量差异表达蛋白,有望成为结肠癌早期诊断的微量蛋白标志物。Objective Analyzing the changes of trace protein profiling in nude mouse models of HCT-116 human colorectal cancer,searching for the unique trace protein markers for the early diagnosis of colorectal cancer. Methods According to the principle of allocation at random,20 of BALB / C-nu,4 weeks-old nude mice were divided into experimental group and con- trol group and 10 in each group, building the nude mouse models of HCT-116 human colorectal cancer and collecting appro- priate blood samples in different times,detecting trace protein profiling of the samples with the PBS Ⅱ/C Protein fingerprint instrument,collecting data automatically and analyzing the expressions of the differentially expressed proteins by using Ci- phergen Proteinehip 3.0 and Biomarker Wizard 3.1 softwares, searching for the tumor-associated differentially expressed proteins. Results There were 120 differentially expressed proteins between experimental group and control group(P〈 0.01) ,but 5 of them had regular changes,the mass-to-charge ratio were 2357Da,2424Da,2637Da,8183Da and 10133Da,in which the 2637Da,8183Da and 10133Da 3 kinds of trace proteins turned out to be enhanced expression gradually after nude mouse had been vaccinated,the 2357Da and 2424Da 2 kinds decreased expression gradually,and that can be learned that the proteins above played very important roles in many aspects such as inducing cell mitosis, enhancing the activity of calcium, participating cell migration and transcriptional regulation and so on by inquiring through the protein database. Conclusion The 5 kinds of trace proteins with regular changes that founded through the study of unique trace protein of the nude mouse models of HCT-116 human colorectal cancer would be expected to become trace protein markers for the early diagnosis of colorectal cancer.
