摘 要:目的探讨用微波方法制备IFN-γ处理的小鼠H22肝癌细胞瘤苗(MITTV-H22)对H22细胞型肝癌的预防与治疗作用。方法 (1)预防实验:采用MITTV-H22免疫小鼠3次后接种H22肿瘤细胞,观察小鼠的生存期、肿瘤生长情况及检测淋巴细胞增殖指数。(2)治疗实验:小鼠接种H22肿瘤细胞的同时接种MITTV-H22进行免疫治疗。观察小鼠的生存期,治疗结束后取瘤称重、测瘤体积。结果 (1)预防实验:实验组小鼠于H22肿瘤细胞攻击后7 d经解剖肿瘤接种部位未发现瘤组织。对照组于接种部位出现直径小于0.5cm的包块,成瘤率为100%。实验组小鼠的生存期(115.40 d±11.22 d vs 31.87 d±5.87 d)和淋巴细胞增殖指数(1.51±0.23 vs 1.26±0.18)都优于对照组(P<0.05)。(2)治疗实验:实验组小鼠的平均生存期长于对照组(46.5 d±3.3 d vs 34.0 d±4.6 d,P<0.05);实验组瘤块体积增长速度明显慢于对照组(F=179.318,P<0.05);实验组的平均瘤体积(0.67 cm^3±0.21cm^3 vs 1.84cm^3±0.74 cm^3)、平均瘤质量(0.67 g±0.35 g vs 1.91 g±0.34 g)小于对照组(P<0.05)。结论 MITTV-H22可以有效的抑制肿瘤生长,具有明显的抗瘤效应。Objective To investigate the prevention and treatment effect on H22 hepatoma tumor vaccine which made by using IFN-γand microwave treated H22 tumor cells ( MITV-H22 ). Methods ( I ) Prophylactic experiment : MITTV-H22 immunized mice 3 times, then H22 tumor cells were inoculated. Tumor growth was observed and spleen cell proliferation was determined by MTT assay. (2)Therapeutic experiment: Mice were inoculated with H22 tumor cells while MITFV-H22 was in- oculated for immunotherapy. The survival period of mice were observed, tumors were weighted and measured. Results ( 1 ) Prophylactic experiment: In experiment group, 7 days after H22 tumor cells challenger, no tumor tissue was found armmd the injection site, even examined with anatomy. In control group, visible tumor (less than 0.5 cm in diameter) at the inoculation site was found with the tumor incidence of 100%. The survival period ( ll5.40 days ± 11. 22 vs. 31.87 days ± 5.87) and proliferation index (1.51 ±0.23 vs. 1.26 -0. 18) were higher in experiment group compared with control group ( P 〈 0.05 ). (2)Therapeutic experiment: The survival period was longer in experiment group compared with control group (46.5 ± 3.3d vs. 34.0 ±4.6d, P 〈0.05) ; The speed of tumor growth in experiment group was slower than that in the control group ( F = 179.318, P 〈 0.05 ) ; The average tumor volume (0.67 cm3 ± 0.21 cm3 vs 1.84 cm3 _± 0.74cm3 ) and tumor weight (0.67 g ± 0.35g vs. 1.91 g -± 0.34g) in experiment group were smaller than that in control group ( P 〈 0.05). Conclusion The MITTV-H22 could effectively inhibit tumor growth and enhance anti-tumor immune response significantly.
