摘  要:目的:观测阿尔茨海默病转基因小鼠不同脑区,比较改良Bielshowsky镀银染色法与免疫组织化学染色法检测老年斑的可靠性。方法:应用体视学方法分别对改良Bielshowsky镀银染色与Aβ免疫组织化学染色结果计数,统计分析两者相关性。结果:Aβ免疫组织化学染色与改良Bielshowsky镀银染色均可检测到AD转基因小鼠不同脑区的老年斑,阳性物计数结果呈正相关性(r=0.228,P<0.01);Aβ免疫组织化学染色法可观察胞质内Aβ的分布。结论:老年斑在AD转基因小鼠不同脑区的分布有差异,改良Bielshowsky镀银染色法与Aβ免疫组织化学染色法均可作为阿尔茨海默病病理特征老年斑的检测手段。Objective To observe senile plaque in the different brain regions of Alzheimer disease(AD) transgenic mice using inprovement Bielshowsky silver staning and immunohistochemistry,and find out the most suitable method by comparing with various experimental results.Methods The stereological metrology was applied in counting the amounts of positive staining objects by inprovement Bielshowsky silver staning and immunohistochemistry,and the two methods correlation was evaluated by statistical analysis.Results The senile plaque in different brain regions of AD transgenic mice?were detected by Aβ immunohistochemistry and improvement Bielshowsky silver staining.There were positive correlation of the counts between the two methods(r=0.228,P0.01),while Aβ distribution in cytoplast was detected with immunohistochemistry.Conclusion There were significant difference of Aβ expression in various brain regions of AD transgenic mice which could be detected by inprovement Bielshowsky silver staning and Aβ immunohistochemistry.
