摘 要:目的探讨TLR4在HPV感染致宫颈病变发展过程中的作用。方法采用RT-PCR法检测TLR4和HPV16 E7在80例HPV阳性的宫颈病变标本(包括正常宫颈,宫颈炎,宫颈上皮内瘤样变和宫颈癌各20例)中的表达。结果随着宫颈病变程度的加重TLR4基因含量逐渐上升,差异有统计学意义(F=16.23,P=0.0041);HPV16 E7的表达也有上升趋势,但差异无统计学意义(F=2.13,P=0.0526)。在宫颈癌中,TLR4的含量和HPV16 E7的含量无明显相关性(γ=0.121,P=0.0612)。结论 TLR4参与了宫颈癌的发生发展,但其在宫颈癌中的表达与HPV16型高危病毒感染无关,即TLR4可能介导了其他病原体感染,从而促进HPV病毒对宫颈上皮细胞的致病作用。Objective To study the role of TLR4 in the occurrence and development of HPV related cervical lesion.Methods RT-PCR method was used to detect the gene expression of TLR4 and HPV16 E7 in 80 specimen of cervical tissue with HPV infection(including normal cervix,cervicitis,cervical intraepithelial neoplasia and cervical cancer,each with 20 cases).Results With the aggravating of cervical lesion,the gene expression of TLR4 was increasing gradully with significant statistical difference(F=16.23,P=0.0041);and HPV16 E7 expression had a tendency to rise up with no significant statistical difference(F= 2.13,P= 0.0526).And there was no obvious correlation between the expression of TLR4 and HPV in cervical cancer(γ=0.121,P= 0.0612).Conclusion LR4 participated in the occurrence and development of cervical cancer,but its role is irrelevant to HPV16.TLR4 maybe mediate another pathogen infection and cause the inflammatory pathologic process due to HPV in cervical epithelial cells.
