摘  要:目的探讨碳酸酐酶Ⅸ(CA-Ⅸ)及缺氧诱导因子-1α(HIF-1α)在前列腺癌不同分期分级中的表达及其内在联系情况。方法采用免疫组织化学S-P法及Western-blot检测正常前列腺组织、前列腺癌组织以及前列腺癌细胞系PC-3、Lncap中CA-Ⅸ及HIF-1α的表达情况,结合临床资料进行统计分析,评价CA-Ⅸ及HIF-1α表达情况与前列腺组织癌变分化程度之间的关系,同时分析两者之间的相关性。结果在正常前列腺组织中CA-Ⅸ及HIF-1α基本不表达,在前列腺癌组织石蜡切片中,HIF处于高表达,其表达情况与前列腺癌病理分级相关。低分化的前列腺癌组织中HIF-1α的表达量高于高分化的前列腺癌组织。CA-Ⅸ在前列腺癌组织中表达率为37.5%,高于正常组织,与肿瘤分化程度无关。CA-Ⅸ及HIF-1α在前列腺癌组织中的表达情况具有相关性。结论 CA-Ⅸ及HIF-1α与前列腺癌的发生成正相关,而且两者在前列腺癌组织中的表达具有相关性,同时提示了以缺氧诱导因子通路为基础的分子机制在前列腺癌的演进中起到一定的作用。Objective To study the expression and correlation of Carbon anhydrase IX (CA-IX) and Hypoxia-inducible factor-1 alpha (HIF-1α) in prostate cancer. Methods The immunohistochemistry of S-P and western-blot were used to detect the expression of Carbon anhydrase IX (CA-IX) and Hypoxia-inducible factor-I alpha (HIF-lct) in normal prostate tissue, prostate cancer tissue, and prostate cancer cell lines PC-3, Lncap. Combined with climcal data, the statistical analysis on the evaluation of CA-IX and HIF-1α expression and prostate tissue differentiation degree relationship was done and the correlation between the two factor was analysed. Results In normal prostate tissue, CA-IX and HIF-1α almost did not express, but in prostate cancer tissue paraffin section, HIF-1α was at a high expression, and its expression had relationship with pathological classification, the lower differentiation of prostatic carcinoma tissues the higher expression of HIF-1α. CA-IX expression rate in prostate cancer tissues was 37.5%, higher than that in normal tissue, and had no relationship with the prostatic tissue differentiation. CA-IX and HIF-1α expression in prostate tissue had correlation, Conclusion CA-IX and HIF-1α had a positive correlation with prostate cancer, and both had correlation with each other in prostate cancer tissue. The hypoxiamolecular mechanism had an important role in the evolution of prostate cancer.
