摘  要:目的:探究大鼠Walker-256移植瘤中Cyclin E的表达水平同荷瘤大鼠接受化疗治疗后生存率的关系。方法:建立大鼠Walker-256移植瘤模型,利用随机表按照安全随机法随机分为4组,对照组、阿霉素组、紫杉醇组、丝裂霉素组。分别以生理盐水、阿霉素组、紫杉醇组、丝裂霉素予以治疗。绘制各组大鼠的生存曲线、并检测各组大鼠移植瘤内Cyclin E的表达水平。以方差分析统计各组之间Cyclin E表达水平的差异,各亚组间荷瘤大鼠的生存率以Wilcoxon法及Kaplan-Meier法进行生存分析。结果:在各组之间,Cyclin E的表达水平无明显差异(P=0.875);在各组中,移植瘤内Cyclin E表达水平高的大鼠生存率较低,反之移植瘤内Cyclin E表达水平低的大鼠生存率较高,除丝裂霉素组外(P=1.000),这种差异有显著的统计学意义(对照组P=0.016,阿霉素组P=0.009,紫杉醇组P=0.043)。结论:CyclinE表达水平的高低同大鼠Walker-256移植瘤对化疗治疗的敏感性有关,对乳腺肿瘤患者进行CyclinE的检测或可有助于预测患者的化疗效果。Objective :To find out the relation- ship between the expression of Cyclin E in Walker -256 tumor bearing rats and the survival of the rats with chemotherapy meth- ods. Methods:We established the Walker - 256 tumor bearing rats model, and divided them into 4 groups randomly: the control group, the adriamycin group, the paelitaxel group and the mitomycin group. We treated the 4 groups with saline, adriamycin, paclita- xel and mitomycin respectively. Then we in- vestigated the survival of the rats and meas-ured the expression of Cyclin E. Then we analyzed the differences of Cyclin E expres- sion between groups with ANOVA, and the survival rate of tumor - bearing rats of sub - groups were analyzed by Wilcoxon methods and Kaplan - Meier methods. Results : There was no significant different of Cyclin E ex- pression between the 4 groups( P = 0. 875 ). In all the groups, the rats which had a high- er Cyclin E expression level had a lower sur- vival rate and the rats which had a lower Cy- clin E expression level had a higher survival rate, and the data have significant differ- ences( control :P = 0. 016, Adriamycin : P = 0.009 ,paclitaxel:P = 0. 043 ) except in mit- omycin group ( P = 1. 000 ). Conclusion : The expression of Cyclin E has a strong relation- ship with the sensitivity to chemotherapy of Walker - 256 tumor bearing rats. The detec- tion of Cyclin E expression might have bene- fit to predict the outcome of chemotherapy to the breast tumor patients.
