摘  要:目的:观察丹龙醒脑方对老年肾虚血管性痴呆大鼠模型海马Bax,Bcl-2蛋白表达及神经元凋亡的影响。方法:用D-半乳糖注射和甲状腺片混悬液灌胃建立老年肾虚大鼠模型,随后行双侧颈总动脉间歇结扎加剪尾放血建立老年肾虚血管性痴呆大鼠模型。采用TUNEL法检测细胞凋亡,免疫组织化学方法检测Bcl-2、Bax蛋白表达。结果:与模型组比较,雌二醇组和丹龙醒脑方组抗凋亡基因Bcl-2蛋白表达显著性增加(P<0.01),凋亡基因Bax蛋白表达降低(P<0.01),凋亡细胞显著性减少(P<0.01)。结论:丹龙醒脑方能通过增强Bcl-2的表达,抑制Bax的表达,抑制神经元凋亡的方式,起到保护神经元,改善痴呆大鼠学习记忆能力的作用,具有和雌激素相似的效应。Objective: To observe the effect of Danlong Xingnao Fang on the hippocampal Bax, Bcl-2 protein expression and neuronal apoptosis of senile kidney deficiency in vascular dementia rat model. Methods: Use D-galactose injection and thyroid tablets suspension gavage to establish senile kidney deficiency rat model, followed by bilateral carotid artery intermittent ligation plus scissors tail bloodletting to establish senile kidney deficiency in vascular dementia rat model. Using terminal deoxynucleotidyl transferase mediated biotin deoxy uridine nucleotide nick end labeling (TUNEL) to detect the apoptotic cells, immunohistochemistry to detect Bcl-2, Bax protein expression. Results: The anti-apoptotic gene Bcl-2 protein expression increased significantly in Estradiol and Danlong Xingnao Fang group(P〈0.01), apoptosis gene Bax expression decreased(P〈0.01),apoptotic cells decreased significantly(P〈0.01). Conclusion: Danlong Xingnao Fang can enhance Bcl-2 expression, inhibit Bax expression and neuronal apoptosis to protect neurons, improve dementia learning and memory ability of rats, it has the similar effect with estrogen.
