摘　　要：目的研究整合素αvβ3抗体对乳腺癌细胞MDA-MB-435S生长的影响,为以整合素αvβ3为靶点治疗乳腺癌提供实验依据。方法采用MTT比色法观察不同浓度整合素αvβ3抗体对体外培养的乳腺癌细胞MDA-MB-435S的杀伤作用,形态学观察细胞变化。结果整合素αvβ3抗体对体外培养的乳腺癌细胞MDA-MB-435S有显著的杀伤作用,其作用表现出明显的量效关系。结论整合素αvβ3对乳腺癌细胞MDA-MB-435S的增殖有促进作用,整合素αvβ3抗体可显著抑制其生长,为抗整合素αvβ3治疗乳腺癌提供了实验依据。Objective To investigate the effect of integrin avβ3 antibody on MDA-MB-435S of the breast carcinoma cell in vitro, so as to provide more experimental bases for breast cancer treatment by anti- avβ3 integrin. Methods The effect of various concentration of integrin avβ3 monoclonal antibody on cell killing was detected by MTT colorinetic method and the changes of cells were observed by morphology. Results Integrin avβ3 monoclonal antibody markedly killed MDA- MB-435S with the breast carcinoma cells in a dose-depended manner. Conclusion Integrin avβ3 can positively regulate the cell malignant proliferation of breast carcinoma, and integrin avβ3 antibody can inhibit it. This result provides more experimental bases for breast cancer by anti-avβ3 integrin.
