摘 要:目的:探讨食管鳞状细胞癌(食管鳞癌)组织中肿瘤相关蛋白表达与临床病理变化的关系。方法:采用免疫组化法检测679例食管鳞癌组织中细胞骨架结合蛋白Fascin、突触蛋白Syn和肿瘤抑制基因P63等23种肿瘤相关蛋白在癌组织中的表达,分析上述蛋白的表达与临床病理指标(淋巴结转移、分化程度及临床分期等)的关系。结果:23种蛋白中,食管癌组织中阳性表达率较高(≥80%)的蛋白是AE1/AE3、P63、CK5/6、CK、Ki67和MUC1,较低(≤30%)的是CD56、CTTN、Syn、S100、CgA和CK20。其中,Syn在淋巴结有转移组织比无转移组织表达升高(χ2=4.167,P=0.041),免疫蛋白CD56在低分化组织高于中高分化组织(P=0.017),细胞增殖抗原Ki67在中晚期组织高于早期(χ2=6.596,P=0.010);而与此相反,P63则在淋巴结无转移时表达上升(χ2=5.095,P=0.024),细胞黏附因子E-cad、Fascin、Ezrin、PLK1在中高分化组织中表达升高(P均<0.05),酪氨酸磷酸化蛋白CTTN和Vim在早期组织高于中晚期组织(P均<0.05)。结论:Syn、CD56和Ki67高表达可能与食管癌预后差有关;而P63、E-cad、Fascin、Ezrin、PLK1和Vim、CTTN高表达可能与食管癌预后好有关。Aim:To detect the expressions of 23 kinds of cancer-related protein and their relation with clinical pathological changes in 679 cases of esophageal squamous cell carcinoma(ESCC),and to further understand the molecular basis for ESCC prognosis.Methods:Six hundred and seventy nine ESCC samples were surgically resected tissue.The expressions of 23 kinds of cancer-related protein,including Fascin, Syn,P63,et al were detected by immunohistochemical method.The relationship between the expressions of 23 kinds of protein and clinical pathological changes was analyzed.Results:Out of the 23 kinds of cancer-related protein,AE1/AE3, P63, CK5/6, CK, Ki67 and MUC1 (≥80%) had relatively higher positive expression rate in ESCC tissue,while CD56, CTTN, Syn, S100, CgA and CK20 (≤30%) had relatively lower positive expression rate.Syn expression was higher in metastasis tissue than in non-metastasis tissue(χ2=4.167,P=0.041),CD56 expression was higher in poorly differentiated tissue than in high and moderate differentiated tissue (P=0.017),and Ki67 expression was higher in advanced stage than in early stage (χ2=6.596,P=0.010). In contrast,P63 expression was higher in ESCC tissue without metastasis (χ2=5.095,P=0.024),the expressions of E-cad, Fascin, Ezrin,and PLK1 were higher in high and moderate differentiated tissue (P0.05),and Vim and CTTN protein expression was higher in early stage than in advanced stage(P0.05).Conclusion:The higher expressions of Syn, CD56 and Ki67 may be important candidates for ESCC with poor prognosis. However, the higher expressions of P63, E-cad, Fascin, Ezrin, PLK1, Vim, and CTTN may be important candidates for ESCC with better prognosis.
