摘  要:目的 :研究香菇多糖对人早幼粒白血病细胞 (HL - 60 )酪氨酸蛋白磷酸化作用的影响。方法 :以不同浓度的药物处理体外培养的HL - 60细胞 ,用放射免疫法测定经过培养 48h的对照组和药物处理组胞浆和膜部分的酪氨酸蛋白激酶 (TPK)和磷酸酪氨酸蛋白磷酸酶 (PTPP)的活力 ,比较酶活力变化情况。结果 :药物对胞浆和膜两部分TPK活力均有一定程度的抑制 ,但抑制作用不明显 (P >0 .0 5) ,而对两部分PTPP活力均有明显的激活作用 (P <0 .0 5或P <0 .0 1 )。结论 :由于上述结果 ,最终导致HL - 60细胞酪氨酸蛋白磷酸化水平下降。Objective:To study the effects of lentinan polysaccharide on tyrosine protein phosphoryaltion in HL-60 cells.Methods:HL-60 cells cultured in vitro were treated by lentinan polysaccharide.The activities of tyrosine protein kinase(TPK) and phosphotyrosyl protein phosphatase(PTPP)in cytosol and membrane fractions were determined,at 48h after administration,by radioimmunoassay ,and the activities of enzymes were analysed between treated and control groups.Results:The difference in activities of TPK between the labor groups and their control groups both in sytosol and membrane fractions was not significant.But the activities of PTPP were increased.Conclusion:on the basis of the aforementioned results,in general,the level of total tyrosine phosphoryaltion was decreased in HL-60 cells,which were treated by lentinan polysaccharide.
