摘 要:为研究荷包猪SLA-3基因特征,设计引物克隆了荷包猪SLA-3,经生物信息学软件分析其基因特征。结果显示,所克隆的荷包猪SLA-3基因(暂命名为SLA-3-HB02)碱基序列长度为1 417 bp,其中2-1 054为ORF区,共编码350个氨基酸,在其编码的氨基酸334-344处存在缺失。SLA-3-HB02与其他SLA-3、SLA-2及SLA-1等位基因群的氨基酸同源率分别为87.8%-98.6%、82.8%-87.8%和83.7%-90.0%。分子进化关系分析,SLA-3-HB02与美国的Hanford遗传距离较近;各功能区分析,SLA-3-HB02保留了HLA-A2等的部分功能基因位点,并与HLA-B15和H-2K1有一定的交叉识别位点。同源模建揭示SLA-3-HB02分子与HLA-A2及H-2K1 3D结构相似。To study the genetic characteristics of Hebao pigs, a pair of primers was designed to clone SLA-3 gene followed by analyzing its genetic characteristics by bioinformatics. The results were shown that the SLA-3 ( named as SLA-3-HB02 )was 1 417 bp with an ORF domain which coded for 350 amino acids and there is a deletion between 334-344 amino acid sites. The amino acid identical ratios between SLA-3- HB02 and other SLA-3, SLA-2, and SLA-1 alleles were 87.8%-98.6%, 82.8%-87.8%, and 83.7%-90.0%, respectively. The phylogenetic tree of the SLA^3-HB02 demonstrated that it was close to Hanford breed of swine in USA. The SLA-3-HB03 preserved some functional sites of HLA-A2 and it also shared some functional sites to the HLA-B 15 and H-2Kd. By homology modeling, the 3-dimentional ( 3D ) structure of SLA- 3-HB02, HLA-A2 and H-2K1 was similar.
