雷利度胺对多发性骨髓瘤KM3细胞的作用机制研究

摘　　要：目的研究雷利度胺对多发性骨髓瘤(MM)细胞KM3细胞凋亡的影响并初步探讨其作用机制。方法用台盼蓝拒染法检测雷利度胺对KM3细胞生长抑制的影响,运用流式细胞仪(Annexin-V/FITC法)检测细胞凋亡。结果不同浓度(0 nmol/L、40 nmol/L、80 nmol/L、100 nmol/L、200nmol/L)的雷利度胺分别作用于KM3细胞24 h后,KM3细胞的存活百分比逐渐减少,分别为100.0%±2.5%,87.2%±1.2%,72.1%±2.4%,61.8%±1.8%,50.1%±0.9%;细胞凋亡率随药物浓度的增加逐渐增加,分别为8.5%±0.8%、11.7%±1.4%、24.7%±1.8%、31.2%±2.3%、35.1%±1.4%。结论雷利度胺抑制KM3细胞的生长并诱导其发生细胞凋亡,而且不同浓度的雷利度胺对KM3细胞呈现剂量依赖性抑制作用。Objective To study the impact of lenalidomide （LEN） on KM3 cels from multiple myeloma patients （MM） and its mecha- nism. Methods The effect of lenalidomide on the KM3 cell growth inhibition was detected by the means of trypan blue staining, and the cells ap- optosis was detected by the means of flow cytometry （ Annexin - V/FITC method）. Results After different concentratios of lenalidomide acted in KM3 ceils for 24hrs, the KM3 cell survival percentage gradually decreased, were 100.0%±2.5% , 87.2% ± 1.2%, 72.1% ± 2.4% , 61.8% ± 1.8 %, 50.1% ± 0.9%, respectively; The apoptosis rate increased gradually with the ratio of drug concentration were 8.5 % ± 0. 8 %, 11.7 % ± 1.4%, 24.7% ± 1.8%, 31.2% ± 2.3%, 35.1% ± 1.4%, respectively. Conclusion Lenalidomide can inhibit the growth of KM3 cells and induce the apoptosis of these cells, hence it plays the role of anti - cancer effect, and shows various degree of dosage dependent inhibition on KM3 cells.
