MMP-3及TIMP-3在人脑胶质瘤中的表达及意义

摘　　要：目的探讨人脑胶质瘤中基质金属蛋白酶-3(MMP-3)及其抑制剂(TIMP-3)的表达及其与恶性程度及侵袭性的关系及临床意义。方法采用免疫组化SP法检测MMP-3和TIMP-3在不同级别胶质瘤患者及正常脑组织中的表达。结果 MMP-3、TIMP-3主要在胶质瘤细胞和血管内皮细胞的胞质表达明显,部分在瘤周较集中。在肿瘤组中MMP-3表达率为72.0%,高于对照组,差异有统计学意义(P<0.05);其表达随胶质瘤级别的增高而增高,二者之间呈正相关。TIMP-3表达率为56.0%,高于对照组,差异有统计学意义(P<0.05);其表达随胶质瘤级别的增高变化不明显,二者之间无明显相关。结论 MMP-3、TIMP-3的表达可能与胶质瘤恶性程度、侵袭性有密切的关系,为临床判断胶质瘤恶性程度、治疗提供一定的指导。Objective To observe the expression of matrix metalloproteinases-3 （MMP-3） and its inhibitors （TIMP-3） in human brain glioma and its relationship with glioma grade and invasion. Methods Used the immunohistochemistry SP methods to detect the expression of MMP-3 and TIMP-3 in glioma and normal brain tissues. Results The MMP-3 and TIMP-3 were obviously expressed in the cytoplasm of glioma and endotheliocyte. Part of them were relatively concentrated in the vicinity of tumor. In the group of tumor,the expression rate of MMP-3 was 72.0% and was higher than the control group, the difference was statistically significant（P 〈 0.05）. Its expression became higher when their malignancy became higher, there was positive correlation between them. The expression rate of TIMP-3 was 56.0% and was higher than was the control group, the difference was statistically significant （P 〈 0.05）. Its expression does not became higher when their malignancy became higher. There was no obvious correlation between them. Conclusion The expression of MMP-3 and TIMP-3 are related with the malignancy and invasion of glioma, may provide some guidance for the malignancy and treatment in clinic.
