摘 要:目的构建一种可靠、有效、实用的毛囊重建模型。方法于6只裸鼠背部分别植入4个直径为2.5mm的微型小室,分离新生C57BL/6小鼠皮肤中表皮和真皮细胞,真皮细胞标记后以1:1比例移植至微型小室内,1周后拆除小室,通过宏观、扫描电镜及组织学观察毛发再生。结果移植后4周,6只裸鼠,24个移植部位均有黑色毛发生长,毛囊重建成功率为100%。冷冻切片荧光检测见毛乳头为红色荧光,石蜡切片苏木素,伊红(HE)染色见毛囊结构发育完整,电镜扫描见毛发生长方向规整,毛发质量优异。结论新生鼠皮肤细胞微型小室法移植后能成功构建毛囊发育模型,此模型具有创伤小、操作简便、所需细胞量少、可做多个部位移植的优点,是一种可靠、有效、实用的毛囊重建模型。Objective To construct a reliable, effective and practical model for hair follicle reconstitution. Methods Six nude mice were transplanted with four mini-chambers with diameter of 2. 5 mm in the dorsal skin respectively. Dermal and epidermal cells were isolated from the skin of newborn C57BL/6 mice. Dermal cells were labeled, then these two kinds of cells were mixed in a ratio of 1:1 and injected into the mini-chambers. Hair regeneration was observed grossly, scanning electron microscopically and histologically. Results Four weeks after cell implantation, black hair grew from the skin in six nude mice and 24 transplantation sites. The successful ratio of hair reconstitution was 100%. Newly formed hair follicles were positive for DiI in the dermal papilla and completely developed structure of hair follicle was observed by HE staining. Good-quality hair grew from the skin regularly was observed under the scanning electron microscopy. Conclusion Newborn mice skin cells were transplanted by using mini-chambers, which can construct a model of hair follicle reconstitution successfully. The model has the advantages of small trauma, simple operation and less cells needed, and several transplantation sites can be made, which is a reliable, effective and practical model for hair follicle reconstitution.
