摘 要:目的研究雌性SD大鼠运动功能及红核NGF及其相关受体表达的增龄性变化。方法选用22月龄(老年组)和3月龄(青年组)SD大鼠,应用网屏握持试验、抓握牵引试验和平衡木试验检测大鼠运动功能;应用West-ern Blot检测红核mNGF、TrkA、Pro-NGF、p75NTR、sortilin的表达。结果老年组大鼠运动功能明显差于青年组大鼠,其前肢力量及平衡感觉明显衰退。与青年组大鼠相比,老年组大鼠红核Pro-NGF、p75NTR、sortilin表达上升,mNGF、TrkA表达下降。结论雌性老年大鼠的衰老伴随着其运动功能及红核mNGF、TrkA表达下降,红核中NGF信号途径的变化和老年大鼠运动功能的衰退相关。Objective To observe the change of motor function and expression of NGF and its receptor in red nucleus of aged female rats. Methods Female Sprague Dawley (SD) rats were divided into two groups. There were aged group ( twenty two - month - old) and young group ( three - month - old). The motor function was evaluated by screen test, prehensile -traction test and balance beam test. Western blot technique was carried out to observe the expression of mature nerve growth factor (mNGF) and its high affinity receptor TrkA, Pro - NGF and its receptor p75NTR,coreceptor sortilin. Results Motor function of aged group was clearly inferior to that of young group. The strength of forelimb and sense of balance in aged group obviously declined. Western blot showed that compared with that of the young rats, the expression of mNGF and TrkA in red nucleus of aged rats was significantly decreased but the expression of Pro - NGF, p75 NTR and sortilin was increased. Conclusion The decline of motor function and the expression of mNGF and TrkA in red nucleus are associated with aging of female rats. The change of NGF signaling pathway in red nucleus is related to the decline of motor function of aged female rats.
