摘　　要：目的：验证利用眼球后静脉注射（retro-orbital injection,ROI）建立小鼠非清髓同种异基因骨髓细胞移植（Non-allogeneic myeloablative bone marrow cell trans-plantation,N-Allo-BMT）模型的可行性和安全性,寻找替代尾静脉注射（tail vein in-jection,TVI）进行小鼠细胞移植的理想途径。方法：对比10名志愿者从练习到掌握ROI与TVI所需时间;经ROI建立N-Allo-BMT模型,利用外周血瑞氏染色及流式细胞计数,连续观测外周血像及嵌合情况,观察移植物抗宿主疾病（GVHD）发生情况。结果：所有志愿者均在1~3h内能熟练完成ROI,且无注射相关并发症发生;在移植10d后,受体小鼠完全恢复造血,外周血呈供体完全嵌合;在移植75d内,83.3%的受体死于严重GVHD。讨论：ROI是一种简便、安全、可靠的小鼠骨髓细胞移植方法,可代替操作难度较大的TVI,值得推广。Object To testify the reliability and safety of establishment of Non-myeloablative allogeneic bone marrow transplantation model（N-allo-BMT） in mice via retro-orbital injection（ROI）,to seek for the ideal replacement of tail vein injection（TVI） for cell transplantation in vivo.Methods 10 participants,who never practiced TVI or ROI,were recruited in this experiment to practice both TVI and ROI,and the total practice time was compared.After transplantation via ROI,consecutive Wright’s staining and FCM（flow cytometry） were performed to observe the hematopoesis and chimerism state.The GVHD in recipients was recorded.Result All of the participants can handle ROI within one to three hours,comparatively,most of participants quite from practicing TVI.There is no injection related complication for all of the recipient mice.10 days after transplantation,all of the recipients restore hematopoiesis,and achieve full donor chimerism.Within 75 days,83.3% of the recipients die from the severe GVHD.Conclusion ROI is much easier to handle,and is a reliable and safe method for bone marrow cell transplantation in vivo.It can be an ideal alternative for the TVI.
