摘 要:生物材料的表面性能是影响材料血液相容性的重要原因 ,生物材料的表面改性对提高其血液相容性有重要意义 ,目前国内外公认的材料表面内皮细胞化是提高生物材料血液相容性最为理想的途径 .作者研究了在PET膜表面通过紫外辐照技术引入丙烯酰胺 ,再通过霍夫曼反应将丙烯酰胺降级为伯胺 ,以提高自由氨基的活性 ,达到了在PET膜材料表面引入氨基酸接口的目的 .然后将保护氨基酸同自由氨基反应 ,将氨基酸以共价键的方式引入到材料表面 .脱保护后用茚三酮检测引入的氨基酸 ,证实了氨基酸在材料表面的存在 .为将来进一步将 (RGD)n共价接枝固定在材料表面 ,使内皮细胞特异识别 ,加速内皮化 ,从而最终提高生物材料的血液相容性 .The major problem of biomaterials is its bad haemo-compatibility of long term using. The biomaterials' surface is the key factor for its haemo compatibility. So, many methods were focused on it. Such as the wetability surface, the negative charge surface and so on. The best way to improve the biomaterials' haemo compatibility is to implant the endothelial cell(EC) onto the surface. For a long period, the oligo peptide RGD were considered is the important recognize site of extracelluar matrix, so do the endothelial cell. The main idea of the research is to introduce the oligo peptide RGD onto the biomaterial surface. The UV irradiation methods was used to graft the acrylamide(AAM) to the surface of the poly(ethylene terephthalate)(PET) firstly. Then the PET films were treated by the Hoffman amide degradation reaction. This step is to active the reaction ability of the -NH 2. Thirdly, the amino acid glycine protected by o-phthalic anhydride was reacted with the PET film. In the forth step, the PET film was treated by the hydrazine hydrate. At last, the PET film was tested by the nihydrine. The positive results showed the glycine was grafted to the surface of the PET film.
