摘  要:目的研究IL-10对β-淀粉样蛋白(Aβ1-42)诱导的阿尔茨海默病(AD)模型大鼠海马淀粉样前体蛋白(APP)表达的影响。方法 48只大鼠随机均分为正常对照组(A组)、PBS对照组(B组)、Aβ1-420.1、0.2、0.4mM注射模型组(分别为C1、C2、C3组)和IL-10 80μg/ml预处理注射组(D组)。注射2d后,Morris水迷宫测定大鼠潜伏期,Western blot法检测海马组织中APP的表达量。结果与A、B组相比,C1、C2、C3组大鼠水迷宫实验潜伏期、APP蛋白表达均呈浓度依赖性的增加(P<0.01);而D组潜伏期、APP蛋白表达较C3组明显减少(P<0.05)。结论海马内注射IL-10能抑制Aβ1-42诱导的AD大鼠水迷宫潜伏期及海马内APP表达的增加,提示IL-10可能抑制由Aβ1-42诱导的AD神经炎症。Objective To investigate the effect of IL-10 on expression of amyloid precursor protein(APP) in the hippocampus of rats with Alzheimerrs disease(AD) induced by Aβ_1-42. Mehtods Forty-eight rats were equally randomized into six groups of A(normal control), B(PBS control), C1 (Aβ_1-42 0. 1 mM),C2 (Aβ_1-42 0. 2 raM), C3 (Aβ_1-42 0.4 mM) and D(IL-10 80μg/ml plus Aβ_1-42 0. 4 mM). Two days after injection, the latent period of rats was measured by Morris water maze, and the protein expression of APP in the hippocampus was detected by Western blot. Results Compared with groups of A and B, the latent period of rats in Morris water maze test and the protein expression of APP were increased in a dose-dependent manner in groups of C1, C2 and C3(P〈0. 01). While the latent period and APP expression in group D were lower than those in group C3(P〈0. 05). Conclusion IL-10 injected into the hippocampus can inhibit the increase of latent period and APP expression in the rats with AD, which indicates that IL-10 may depress the nerve inflammation of AD induced by Aβ_1- 42.
