摘 要:目的 观察多西他赛对兔角膜碱烧伤新生学管化及角膜组织中血管内皮生长因子(vascular endothelial growth factor VEGF)的表达影响,评价多西他赛对兔碱烧伤后角膜新生血札管的抑制作用并初步探讨其作用机制,为该药的临床应用提供实验依据,从而为药物治疗角膜新生血管提供一条新途径,方法健康新西兰白兔24只(南昌大学实验动物科学部提供),体质量1.5-2.0Kg,雌雄兼有,双眼均为实验眼,动物实验遵循国家科学技术委员会颁布的《实验动物管理条例》,碱烧伤后兼有随机将24只兔分为对照组和3个实验组,选择A、B、C组多西他赛滴眼液浓度分别为10μg.ml^-1、20μg.ml^-1、40μg.ml^-1,每组6只12眼,对照组D组滴NS眼液每日3次作为阴性对照,共观察14d。所有动物均于碱烧伤后24h开始,每日4次给予氧氟沙星滴眼液眼预防感染,连点三日。应用显微镜,角膜照相观察并测量第7,7,14d各组CNV长度,利用图像分析系统计算各组CNV的面积。取角膜组织行病理检查、免疫组织化学半定量观察及反转录酶-聚合酶连锁反应(reverse transcripton-polymerase chain reacton,RT-PCR)检查VEGF及VEGF mRNA(Messenger RNA,信使核糖核酸)表达等方法,研究角膜新生血管的形成及抑制情况。结果(1)裂隙灯显微镜观察结果:治疗组A、B、C组CNV生长明显受抑制,与对照组比较,新生血管发生时间较迟、生长缓慢、新生血管化面积小,治疗组B、C组兔角膜新生血管面积低于治疗组A和对照组,差异显著有统计意义(P〈0.05),B、C组间差异无统计学意义(P〉0.05).(2)免疫组化染色显示治疗组B、C组VEGF的表达较治疗组A和对照组明显减少,RT-PCR在造模后各时间点角膜组织中VEGF mRNA的相对表达量,治疗组B、C组低于治疗组A和对照组,差异有统计学意义(P〈0.05)。结论多西他赛滴眼液可有效抑制�Objective To evaluate the effect of Docetaxel eye drops on cornea neovasculariza- tion of rabbit induced by alkali cauterization. Methods Cornea neovascularization (CNV) was in- duced by 1 mol.l' NaOH in 24 rabbits. The rabbits were then divided into four groups, group A: 10ug.ml' Docetaxel treated group (n=6), group B: 20ug.ml~ Docetaxel treated group (n=6), group C: 40 ug.ml^-1 Docetaxel treated group (n=6), group D: control group (n=6), group A, B and C h^d been given the eye drops since the 2 days after alkali injury. The eyes in each group were exam- ined by microscope at 3, 7, 14 days. The area of CNV was marked by photography. Immunohisto- chemistry and RT-PCR were used to determine the VEGF level. Results Docetaxel eye drops both inhibited the length and area of rabbit corneal neovascularization at different concentration. However,there was a little difference between the three concentrations of Docetaxel eye drops. The expression of VEGF was suppressed significantly in Docetaxel eye drops group, nevertheless the expression of VEGF was markedly increased in the control group. Conclusions Docetaxel is effective in the inhi- bition of corneal neovacularization induced by alkaline burn.
