摘  要:目的探讨细胞滴片质量对荧光原位杂交(FISH)技术诊断膀胱癌的的影响。方法采用3、7、17号染色体着丝粒探针和9号染色体p16基因位点探针对45例膀胱移行细胞癌患者和50名健康人群的新鲜尿液进行FISH检测,分析细胞滴片对FISH结果的影响。结果标准的尿液脱落细胞滴片为细胞均匀分布,细胞核轮廓清晰,背景干净,红、绿信号清晰明亮。细胞数量太多、细胞间黏液太多、大量的中性粒细胞和淋巴细胞混杂在脱落的上皮细胞中、尿液中含有大量的杆菌、球菌和精子都会导致细胞重叠或探针无法杂交到目的基因上,红、绿信号模糊不清,影响FISH结果的可靠性。结论细胞滴片的质量直接影响到FISH结果的可靠性。Objective To investigate the effect of spinspot on the diagnosis of bladder cancer from voided urine with fluorescence in situ hybridization (FISH) technique. Methods We detected the fresh urine samples from 45 patients with bladder transitional cell cancer and 50 healthy people by centromeric probes for chromosomes 3, 7, 17 and chromosome 9 gene pl6 probe in FISH test, and analyzed the effect of spinspot on FISH results. Results The standard spinspot of urine exfoliated cells had even distribution, with clear nucleus contour, clean background, clear and bright red, green signals. Too many cells, too much intercellular myxoid, large numbers of neutrophils and lymphocytes mingled with the shedding epithelium cells, large amounts of coli, staphylococcus aureus and sperms in urine would lead to cell overlap or the probe not binding to target gene, with obscure red and green signals, and unreliable FISH results. Conclusions The quality of spinspot directly affects the reliability ofFISH results.
