四硫化四砷对人卵巢癌裸鼠皮下移植瘤抑瘤作用的研究

摘　　要：目的探讨四硫化四砷（As4S4）对卵巢癌裸鼠皮下移植瘤的生长和瘤细胞凋亡的影响。方法建立卵巢癌SKOV3裸鼠皮下移植瘤模型，观察As4S4对肿瘤生长的抑制情况；RT—PCR法检测As4S4对卵巢癌裸鼠皮下移植瘤组织中bcl-2和baxmRNA表达水平的影响；Westernblot法检测As4S4对卵巢癌裸鼠皮下移植瘤组织中bcl—2和bax蛋白表达的影响。结果As4S4作用于卵巢癌裸鼠皮下移植瘤后，肿瘤的体积和重量明显减小，而且随药物剂量的增加抑瘤率明显增加。As4S4作用后，RT—PCR可见bcl-2mRNA表达强度减弱，baxmRNA表达强度增强；Westernblot法可见bcl-2蛋白带变细，bax蛋白带增粗。结论As4S4可以抑制卵巢癌裸鼠皮下移植瘤的生长，诱导肿瘤细胞凋亡，其作用机制可能与调节Bcl-2／Bax的表达有关。Objective To investigate the inhibitory effect of As4 S4 on growth and apoptosis of the human ovarian cancer subcutaneously transplanted in nude mice. Methods We established the subcutaneously trans- plantated tumor of SKOV3 by BALB/C nude mice, and evaluated the inhibitory effect of As4 S4 on the growth of tumor tissue;By RT -PCR, we evaluated the influence of As4 S4 on the expression of Bcl -2/Bax mRNA in tumor tissue;By Westemblot, we evaluated the influence of As4S4 on the expression of Bcl -2/Bax protein in tumor tissue. Results After subcutaneously transplanted tumor in nude mice, it was treated with different concentration of As4S4. It was significantly smaller in size and weight. The expression of Bcl - 2 was down - regulated in As4 S4 treated group,while Bax was up - regulated. Conclusion As4S4 suppresses the growth of implanted tumor in nude mice,and it induces tumor cells apoptosis. Anti -tumor activities of As4S4 might be related with inducing apoptosis and Bcl- 2/Bax expression change.
