摘  要:目的观察实验性变态反应性脑脊髓炎(EAE)大鼠正中隆起(ME)在不同发病时期室管膜细胞的变化。方法将20只健康雌性Wistar大鼠随机分为实验组15只和对照组5只,实验组以豚鼠脊髓匀浆加完全福氏佐剂制备免疫抗原,注射于大鼠四肢足垫内,建立EAE模型;对照组四肢足垫内注射生理盐水。实验组于注射后7 d(潜伏期)、14 d(发病极期)、21 d(恢复期)分别随机选择5只处死,对照组于实验21 d处死,采用光镜和透射电镜观察实验组不同时期及对照组ME室管膜细胞的形态改变。结果光镜下对照组室管膜带室管膜细胞排列整齐,呈单层或双层,较稀疏;实验组7 d室管膜带细胞数目增多,14 d达到高峰,21 d基本恢复正常;透射电镜下对照组室管膜细胞脑室面可见微绒毛和微突起,核染色质疏松,细胞之间可见紧密连接;实验组7、14 d室管膜细胞分泌功能旺盛,21 d基本恢复正常。结论 ME在EAE时发生病理改变早于临床症状,且与病情进展程度呈正相关,提示其可能参与脑的免疫调节。Objective To observe the changes of ependymal ceils of median eminence during different stages of experimental allergic encephalomyelitis (EAE) rats. Methods Twenty healthy female Wistar rats were randomly divided into experimental group (n = 15 ) and control group (n = 5). Immunizing antigen made from both complete Freund adjuvant and spinal cord homogenate of guinea pig were injected into rats'paw pads in the experimental group to induce EAE, normal saline were injected into rats'paw pads in the control group. After injected for 7 d (the latency period), 14 d (peak of the diseased period) and 21 d (the recovery stage) , 5 rats of each stage of the experimental group were randomly selected and fixed with cardiac perfusion. Rats of the control group were fixed with cardiac perfusion at 21 d. Then the morphological changes of ME ependymal cells in different stages of experimental group and control group were observed by light microscopy and transmission electron microscope (TEM). Results Light microscope observation showed that the ependymal cells in ependymal zone of the control group were one or two layers, sparse and scattered neatly; the number of ependyma cells in ME of experimental group rats increased at 7 d, reached the peak at 14 d, and reduced near to normal level at 21 d. TEM observation showed there were microvilli and micro projections on the ventricular surface of the ependymal cells in the control group, those ependymal cells had loose chromatin in cell nucleus and tight junction between each other; in experi- mental group, secretory founction of the ependymal cells in ME were vigorous at 7 d and 14 d, reduced near to normal at 21 d. Conclusion The morphological changes of the ME are prior to clinical symptoms and consistent with the progression of EAE, which indicate that the ME is involved in cerebral immunoregulation during EAE.
