摘 要:目的通过在体实验观察高同型半胱氨酸血症动物学习记忆功能及海马长时程增强(LTP)的变化,探讨补充叶酸是否能够改善高同型半胱氨酸血症所导致的学习记忆改变。方法以腹腔注射同型半胱氨酸(Hcy)的方法建立高同型半胱氨酸血症动物模型,动物分为对照组、Hcy处理组、Hcy处理补充叶酸组和补充叶酸组。结果动物经过四周腹腔内注射Hcy,血浆Hcy水平异常升高,动物的生长受到明显抑制。实验动物在注射Hcy后对外界环境的变化恐惧心理增强,自发探究行为减少,动物出现明显的学习记忆障碍。腹腔注射Hcy大鼠海马突触传递效率显著降低。补充叶酸大鼠的学习记忆能力及LTP与对照组无显著差别。结论 Hcy可使动物自发探究行为减少,出现明显的学习记忆障碍,LTP的改变可能是机制之一。补充叶酸可以显著改善高同型半胱氨酸血症所导致的学习记忆功能改变。Objective To observe the effects of hyperhomocysteinemia on the behavior and ability of learning and memory and after folate intervention.Method The Wistar rats were randomly assigned to four groups as follows: control(C),hpyerhomocysteinemia(H),hpyerhomocysteinemia with folate supplement(HF) and folate supplement(F).The hpyerhomocysteinemia will be established by i.p.injection of homocysteine(Hcy).The rats’ spontaneous exploratory behavior and the learning and memory function were detected through open field test and water maze test after 4 w treatment.Nerve electrophysiology of hyperhomocysteinemia rats was detected through long-time potential(LTP).Results After intraperitoneal injection of Hcy the rats’ spontaneous exploratory behavior decreased and the ability of learning and memory was disordered.Compared with C,HF and F groups,the amplitude of LTP in hippocampal dentate gyrus in H group decreased significantly.Folate supplement could inhibit the changes of behavior,the learning and memory function,and LTP induced by Hcy.Conclusion Hyperhomocysteinemia may disorder the ability of learning and memory.Folate could significantly repress the Hcy-induced learning and memory function damage.
