摘 要:目的建立大鼠角膜芥子气损伤的动物模型,探讨鼠角膜芥子气轻度损伤的组织形态学变化。方法设研究组20只,对照组20只,正常组8只。研究组和对照组分别用400μl/L(0,2ml)芥子气液和丙二醇(0.2ml)接触角膜2min,然后于15min,6h.24h,48h,7d,取左眼角膜组织,行HE染色,光镜下进行比较分析。结果15min角膜袁层鳞状上皮脱落,呈毛鳞片状外观。角膜基质层胶原轻度水肿,可见少量中性粒细胞聚集。6h角膜表层鳞状上皮大部分脱落,上皮层变薄。基质层胶原显著疏松,中性粒细胞减少。24h角膜上皮增生、增厚,细胞复层化明显,中性粒细胞明显减少。8h角膜上皮层次增多,毛鳞片状外观消失,表面光滑,偶见凋亡细胞。基质层胶原疏松减轻,偶见中性粒细胞。7d角膜上皮层增厚,偶见凋亡细胞。基质层胶原疏松明显减轻,未见中性粒细胞。对照组角、膜组织正常。结论低浓度的芥子气液可致鼠角膜轻度损伤,出现一过性角膜上皮层和基质层组织形态学重塑改变。Objective To investigate histomorphology development process of mustard-gas-induced rat corneal mild lesion by es- tablishing the animal model. Methods Collect 20 spec, imens of rat corneas injured by liquid mustard gas with the density of 400t, I/L as ex- perimental group,20 specimens injured by propanediol as contronl group and 8 heahhy specimens as normal group. All the specimens were observed with H Ematine staining and by light microscopy. Results Corneal epithelium fell off after 15rain contacting with liquid mustard gas and presented hair scaly-like structure. The collagen in corneal stroma layer became miht edema and few neutrophilic grauulocyte(NG) aggre- gated. After 6h most of the epithelial la,nina fell off,corneal stroma layer collagen was loosen,edenm became notable and quantity of NG de- creased. After 24h corneal epithelium became proliferating and thickening. After 48h and 7 days corneal epithelium layer grew in number and hair scaly-like stnuqure disappeared,as well apoptotie cell was observed by chance. Conclusion Low concentration of mustard gas can cause mild pathological lesion of the rat corneal and transient histomorphology remodeling in corneal epithelium and stroma can be observed.
