摘  要:[目的]探讨TS、ERCC1和DNA-PKcs在大肠癌组织中的表达及其临床意义。[方法]将133例大肠癌标本与17例正常大肠组织标本构建成组织芯片,应用免疫组化SP法检测TS、ERCC1和DNA-PKcs在大肠癌芯片中的表达,并分析其与大肠癌临床病理特征的关系。[结果]TS在大肠癌组织中的表达显著高于正常大肠组织(P<0.05),而ERCC1、DNA-PKcs在大肠癌组织中的表达与正常大肠组织无明显差异(P>0.05)。TS、ERCC1和DNA-PKcs的表达均与淋巴结转移和Dukes分期有关(P<0.05)。ERCC1和DNA-PKcs、TS和ERCC1表达间均呈明显正相关(P<0.01)。[结论]检测TS、ERCC1和DNA-PKcs在大肠癌中的表达对预测大肠癌的发生、发展有一定意义。[Purpose] To investigate the expression of thymidylate synthase(TS), excision repair cross-complementing-1(ERCC1)and DNA-dependent protein kinase catalytic subunit (DNA-PKcs) in colorectal carcinoma and its clinical significance. [Methods] A tissue microarray was constructed from 133 cases colorectal carcinoma and 17 cases with normal colorectal specimens, and the expression of TS,ERCC1 and DNA-PKcs were examined by immunohistochemistry SP method. The relationship of their expression with clinicopathological features of colorectal carcinoma was analyzed. [Results] The positive rate of TS in colorectal carcinoma was significantly higher than that in normal colorectal tissues(P0.05), while the positive rate of ERCC1 and DNA-PKcs in colorectal carcinoma were all similar to those in normal colorectal tissues (P0.05). The expressions of TS,ERCC1 and DNA-PKcs in colorectal carcinoma were all related to lymph node metastasis and Dukes staging (P0.05). There was a positive relationship between the expression of ERCC1 and DNA-PKcs as well as TS and ERCC1(P0.05).[Conclusion] Detecting the expression of TS, ERCC1 and DNA-PKcs in colorectum might predict the prognosis of colorectal carcinoma.
