摘　　要：[目的]对早期和晚期初诊鼻咽癌患者肿瘤组织在蛋白质组水平上的表达差异进行比较研究,识别和鉴定差异蛋白,并对其功能进行初步了解。[方法]按照临床分期将所收集鼻咽癌组织分为早期患者组(I、II期)和中晚期患者组(III、IV期),分别提取两组组织总蛋白,进行双向凝胶电泳(two-dimensional difference gel electrophoresis)分离,利用UMAX PowerLook1100型多功能透射扫描仪不同波长的激光扫描后,获取凝胶图像,应用PDQuest7.1.0软件包进行图像分析,切取凝胶中差异表达≥1.5倍(P≤0.001)的蛋白质斑点进行胶内酶解,应用基质辅助激光解吸/电离-飞行时间质谱仪(MALDI-TOF/MS)进行蛋白质的肽指纹图谱鉴定。[结果]翻译起始因子亚基eIF3K和eIF5A在中晚期鼻咽癌中表达丰度升高。[结论]早期和晚期鼻咽癌组织存在表达差异的蛋白,翻译起始因子亚基eIF3K和eIF5A在早期和中晚期鼻咽癌组织中的差异将为解析鼻咽癌发生发展机制提供线索,也为筛选具体潜在价值的肿瘤化疗靶位点提供理论依据。[Objective]Protein plays an important role in the development of tumor.To identify new related protein,to compare protein differences of tumor tissue between early stage and advanced stage of nasopharyngeal carcinoma.[Mehtods]Patients were divided into 2 groups based on the clinical stages,group A（stage I and II）and group B（stage III and IV）,total protein was then abstracted from tumor tissues in each group for two-dimensional gel electrophoresis separation,the gel image was obtained by laser scanner and analysed with PDQuest 7.1.0 packages.Two significantly changed protein spots were chosen for analysis of protein peptide fingerprint by using MALDI-TOF/MS mass spectrograph.[Results]eIF3,eIF5A increased at early stage and advanced stage of nasopharyngeal carcinoma.[Conclusion]Expression difference of protein is found between different stages of nasopharyngeal cancer.eIF3K and eIF5A as subunits of translation initiation factor may help to clarify development mechanism of nasopharyngeal carcinoma and provide theoretical foundation for screening the promising target positions of tumor chemotherapy.
