摘 要:[目的]建立代谢综合征动物模型并观察其靶器官相关改变。[方法]36只雄性SPF级3周龄雄性Wis-tar大鼠,随机分成3组:正常盐组(NS,n=12)、高盐组(HS,n=12)及高盐高脂组(HSF,n=12)。实验结束,测量体重、颈总动脉有创血压、血钾、血钠、血糖(FBG)、血脂、胰岛素(INS)浓度及胰岛素抵抗指数(HOMA-IR)、病理学检查胸主动脉内膜增厚指标、胰岛素表达以及胰岛β细胞凋亡检测。[结果]实验结束时,高盐高脂组大鼠表现出典型MS特征并观察到胸主动脉内膜增厚、胰腺组织的β细胞表达下降以及胰腺β细胞凋亡等改变。(1)体重增加:HSF组体重明显增加,与NS及HS组比较,P﹤0.05。(2)血压升高:HSF组收缩压均明显升高,与NS组比较,P﹤0.01;HSF组与HS组之间无明显差异。(3)血脂异常:HSF组较NS组大鼠血脂明显增高,以TC、TG、LDL-C为主,三者均P﹤0.05,HDL-C无明显改变(P﹥0.05),HS组与NS组之间无明显差异。(4)胰岛素抵抗:HFS组大鼠胰岛素抵抗指数较NS组明显升高(P﹤0.01),HS组与NS组之间无明显差异;(5)胸主动脉内膜增厚指数(I/M):HSF组及HS组I/M比值高于NS组(0.219±0.050),P﹤0.05;HSF组I/M比值高于HS组,P﹤0.05。(6)免疫组化检测提示与NS组比较,HFS组大鼠胰腺组织的β细胞表达下降。(7)TUNEL胰岛β细胞凋亡检测提示与NS组比较,HFS组细胞凋亡明显增加。[结论]实验结果提示,通过高盐高脂饲料喂养3周龄雄性Wistar大鼠能够诱导形成MS模型,并且可以观察到HFS组大鼠胸主动脉内膜增厚及其胰腺组织β细胞表达下降、胰岛β细胞凋亡明显增加等改变。[Objective]To establish an animal model of metabolic syndrome and observe the changes of critical organs.[Methods]36 male specified-pathogens free(SPF)Wistar rats,aged 3 weeks,were divided into three groups in randomized manner:(1)control with normal salt diet(0.5% NaCl,NS group);(2)high salt diet(4% NaCl,HS group);(3)fat and salt-enriched diet(49%fat+4%NaCl,HSF group),12 animals in each group.Then the bodyweights were measured,the right carotid artery was calculated for direct blood pressure measurement;detected blood glucose,blood-fat,concentration of Insulin(INS),insulin resistance index(HOMA-IR),thickening of tunica intima of aorta pectoralis by pathology,islet insulin expression and apoptosis of beta cell by enzyme linked immunosorbent sorbent assay(ELISA).[Results](1)Body weight significantly increased in HSF group,compared with NS group and HS group(P﹤0.050);(2)The systolic pressure in HSF group significantly improved compared with NS group(P﹤0.01),was not different with HS group;(3)Comparing with NS group,TC,TG,LDL-C in HSF group significantly increased(P﹤0.05),but HDL-C was not significantly different(P﹥0.05);and TC,TG,LDL-C and HDL-C in HSF group were not significantly different with HS group;(4)Insulin resistance index was higher in HSF group than in NS group(P﹤0.01),but there was no significant difference in HS group and NS group;(5)Thickening index(I/M)of tunica intima of aorta pectoralis was higher in and HSF and HS groups than NS group,I/M in HSF group was higher than in HS group(P﹤0.05);(6)beta cell of islet insulin expression in HSF group decreased more than NS group;(7)Cell apoptosis of beta cell in HSF group obviously increased compared with NS group.[Conclusion]The fat and salt-enriched diet given to rats could induce metabolic syndrome,which corresponds or is analogous to the clinical metabolic syndrome;and it also leads to thickening of tunica intima of aorta pectoralis,decrease of
