摘 要:建立杜仲叶多成分的HPLC含量测定方法。建立的色谱系统为:用C18色谱柱,以含0.1%甲酸的乙腈和水为流动相,柱温30℃,波长270 nm,流速1 mL/min。通过对杜仲叶中7种成分(绿原酸、槲皮素-3-O-α-L-吡喃阿拉伯糖基-(1→2)-β-D-葡萄糖基苷、芦丁、异槲皮苷、紫云英苷、槲皮素、山奈酚)的含量测定,确定药材质量的优劣。本方法可使7种成分有效分离,在各自的检测范围内线性良好,精密度和重复性良好,加样回收率范围在99.03%到107.1%之间。经对10批次杜仲叶的测定,认为该方法可以准确地反映杜仲叶质量的优劣。A high-performance liquid chromatographic method was established to evaluate the quality of Eucommiae Folium with a simultaneous determination of seven compounds,chlorogenic acid,quercetin 3-O-α-L-arabinopyranosyl-(1→2)-β-D-glucopyranoside,rutin,isoquercitrin,astragaline,quercetin and kaempferol.The samples were separated on a Hedera-C18 column with gradient elution of acetonitrile and 0.1% methanoic acid at a flow rate of 1.0 mL/min and detected at 270 nm.The seven compounds were completely separated within 45 min and the determinations indicated good linearity.The average recoveries varied from 99.03% to 107.05%.This method was validated with respect to precision,repeatability and accuracy,and was successfully applied to the quantification of seven compounds in 10 commercial samples of Eucommia ulmoides Oliv..
