摘  要:目的检测人胰腺癌组织中DNA甲基化转移酶3b(DNMT3b)的表达,分析其与肿瘤临床病理特征的相关性。方法应用蛋白质印迹法检测12例配对人胰腺癌和癌旁组织中DNMT3b的表达;免疫组织化学法检测59例胰腺癌组织中DNMT3b的表达,并分析其与肿瘤临床病理特征的相关性。结果蛋白质印迹法结果显示,胰腺癌及癌旁组织DNMT3b蛋白表达量分别为0.69±0.13和0.14±0.03,癌组织的表达量明显高于癌旁组织(t=4.464,P〈0.05)。免疫组化结果显示,胰腺癌组织DNMT3b阳性高表达率为59%,癌旁组织阴性表达。肿瘤组织DNMT3b表达强度与TNM分期和淋巴结转移相关。结论DNMT3b在胰腺癌组织中高表达,其表达与肿瘤的恶性生物行为有关。Objective To investigate the expression of DNMT3b and its correlation with clinicopathological parameters of pancreatic cancer. Methods The expressions of DNMT3b protein in 12 pancreatic cancer tissues and para-cancerous tissues were detected by Western blot. The expressions of DNMT3b protein in 59 pancreatic cancer tissues were detected by immunohistochemistry. The association of DNMT3b expression and clinicopathological parameters of pancreatic carcinoma were analyzed. Results Western blot results showed the expressions of DNMT3b protein in pancreatic cancer tissues and para-cancerous tissues were 0.69 ± 0.13and 0.14± 0.03, and the protein level of DNMT3b in pancreatic cancer tissues were significantly higher than that in para-cancerous tissues (t = 4. 464, P 〈0.05 ). Immunohistochemistry examination showed that the positive rates of DNMT3b protein were 59% in pancreatic cancer tissues and negative in para-cancerous tissues. DNMT3b expression was positively correlated with the TNM stage and lymph node metastasis. Conclusions DNMT3b is highly expressed in pancreatic cancer tissues, and it is related with malignant biological behaviors of oancreatic cancer cells.
