Hec1在胃癌组织中的表达及其在胃癌进展中的临床意义

摘　　要：目的:Hec1(癌高表达蛋白),作为着丝粒蛋白的重要组分及纺锤体检测点调节器,在肿瘤的发生和进展中发挥着重要的作用。因此,本文旨在研究Hec1在胃癌组织中的表达及其在胃癌进展中的临床意义。方法:采用Sybr Green荧光染料法的实时定量逆转录PCR方法检测30例胃癌标本及17例癌旁正常组织中Hec1 mRNA表达情况,其中TBP作为内参照。通过2-△CT法计算经过内源性管家基因标准化处理后的目的基因的表达,以此来比较各比较组之间Hec1 mRNA的表达差异。结果:(1)胃癌组织中Hec1 mRNA的表达(0.272±0.0369)明显高于癌旁正常组织(0.041±0.0231)。(2)Ⅰ/Ⅱ期(0.286±0.1843)、Ⅲ/Ⅳ期(0.269±0.0717)胃癌组织Hec1 mRNA表达量明显高于癌旁正常组织;但Ⅰ/Ⅱ期与Ⅲ/Ⅳ期胃癌组织相比较,Hec1 mRNA表达量没有明显差异。(3)Hec1 mRNA在无淋巴结转移组中的表达为0.325±0.2077,在有淋巴结转移组中的表达为0.238±0.0697;两者间无明显差异。结论:Hec1 mRNA在胃癌组织中的表达明显高于癌旁正常组织,但与TNM分期及淋巴结转移情况无明显的相关性。Objective Hec1（highly expressed in cancer）,as a component of kinetochore protein and spindle checkpoint regulator,plays an important role in cancer progression.In this article,we exploredthe expression of Hec1 mRNA and the significant clinical value in gastric cancer.Methods The mRNA expression levels of Hec1 in 30 gastric cancer tissues and 17 corresponding normal tissues were measured by real-time quantitative PCR with flourescent dye SYBR Green,the expression of TBP as internal control.The difference of Hec1 mRNA expression between the two groups was compared in relative quantitative value,which was calculated by mean of 2-△CT.Results（1）The Hec1 mRNA expression was obviously higher in gastric cancer tissues（0.272 ± 0.036 9） than the corresponding normal tissues（0.041 ± 0.023 1）.（2）The Hec1 mRNA expression in gastric cancer grade Ⅰ/Ⅱ（0.286 ± 0.184 3）and Ⅲ/Ⅳ（0.269 ± 0.071 7）is much higher than the corresponding normal tissues,but there is no difference between grade Ⅰ/Ⅱ and Ⅲ/Ⅳ.（3）The Hec1 mRNA expression is 0.325 ± 0.207 7 in group of cancer with no lymph nodes metastasis,0.238 ± 0.069 7 in group of cancer with lymph nodes metastasis;There is no difference between them.Conclusions The Hec1 mRNA expression is obviously higher in gastric cancer tissues than the corresponding normal tissues,but there is no evident correlation with TNM and lymph nodes metastasis.
