摘  要:目的探讨代谢型谷氨酸受体5在甲醛诱导大鼠颞下颌关节疼痛中的作用。方法 SD大鼠36只,随机均分为6组:正常对照组(不做任何处理)、生理盐水组(右侧颞下颌关节内注射50μL生理盐水)、甲醛组(右侧颞下颌关节内注射50μL5%甲醛)、MPEP-1组[25μL MPEP(1 mmoL/L)+50μL 5%甲醛]、MPEP-2组[25μL MPEP(2.5 mmoL/L)+50μL 5%甲醛]、MPEP-3组[25μL MPEP(5 mmoL/L)+50μL 5%甲醛]。将5%甲醛注入右侧颞下颌关节腔内,诱导大鼠颞下颌关节疼痛,计数各组大鼠60 min内伤害性行为反应(包括刮擦头面部及缩头),分光光度计测量颞下颌关节内Evan′s蓝结合血浆蛋白渗出量的变化。结果甲醛组大鼠搔刮头面部及缩头反应持续时间明显高于正常对照组及生理盐水组(P<0.05),MPEP-3组大鼠搔刮头面部及缩头等伤害性行为反应明显低于甲醛组,差异有统计学意义(P<0.05)。甲醛组颞下颌关节内Evan′s蓝结合血浆蛋白渗出量明显高于正常对照组及生理盐水组(P<0.001),MPEP-3组大鼠颞下颌关节内Evan′s蓝结合血浆蛋白渗出量明显低于甲醛组(P<0.05)。结论代谢型谷氨酸受体5介导甲醛诱导大鼠颞下颌关节痛,其拮抗剂能减轻甲醛引起的颞下颌关节疼痛。Objective To explore the function of metabtrophic glutamate receptor subtype 5(mGluR5) in formalin-induced temporomandibular joint pain.Methods A total of 36 adult SD rats were randomly divided into 6 groups(n =6),including normal control group(no-treated),negative control group(Nacl 0.9% injection),formalin group(5% formalin injection),MPEP-1 group(5% formalin +1 mmoL/L of MPEP injection),MPEP-2 group(5% formalin +2.5 mmoL/L MPEP injection),MPEP-3 group(5% formalin +5 mmoL/L MPEP injection).The nociceptive behavioral responses characterized by rubbing the orofacial region and flinching the head quickly were quantified for 60 min.Plasma extravasation of Evan′s blue dye bound to the plasma protein were measured.Results The behaviors characterized by flinching the head and rubbing the orofacial region in formalin group were significantly increased compared with control(P 0.05).The nociceptive behavior in MPEP-3 group was reduced than that in formalin group(P 0.05).Compared with control group,plasma extravasation of Evan′s blue dye bound to the plasma protein of MPEP groups and the formalin group increased significantly(P 0.05);There were statistically significant difference between formalin group and MPEP-3 group(P 0.05).Conclusion Metabtrophic glutamate receptor subtype 5 was involved in formalin-induced temporomandibular joint pain.Metabtrophic glutamate receptor subtype 5 antagonist(MPEP) could alleviated formalin-induced temporomandibular joint pain.
