摘 要:研究和发展了基于表面等离子体共振(Surface Plasmon Resonance,SPR)生物传感器进行牛奶中抗生素残留检测的信号放大策略。在竞争法检测的基础上,采用二次抗体进行信号放大,提高传感器检测的灵敏度,降低检测限。并对两种抗生素残留物链霉素、卡那霉素的水溶液进行了实验。结果表明,利用竞争法检测,两种残留物的最低检测限(Limit of Detection,LOD)分别为34 ng/mL和25 ng/mL,而利用信号放大法检测,两种残留物的最低检测限LOD降低为24 ng/mL和12 ng/mL。该方法丰富了SPR生物传感器检测抗生素等小分子残留物的理论。Signal amplification assay based on the surface plasmon resonance (SPR) biosensor for the detection of antibiotics residue in milk is developed in this paper. In an attempt lower limit of detection (LOD), second antibodies were introduced competitive inhibition assay. The second analyte can magnify the signals to improve the sensitivity and get more for second detecting analyte after the obviously made by the first detection of the analyte. The detection experiments of streptomycin and kanamycin in water solution were carried out. The results show that the LODs of the residues using the competitive inhibition assays are 34 ng/mL and 25 ng/mL, respectively, while the LODs using the signal amplification assays are 24 ng/mL and 12 ng/mL, respectively. So the signal amplification detection not only satisfies the regulation of EU' s maximum residue hmit (MRL) , but also is better than the competitive inhibition method. The study has developed and enriched the theory of detecting low molecular residues by using SPR biosensor.
