摘　　要：目的探讨长春新碱耐药的人类胃癌细胞株SGC7901/VCR发生多药耐受(MDR)的机制。方法采用比较蛋白质组学方法、反义寡核苷酸(ASOs)抑制法、质谱分析(MS)和免疫共沉淀(IP)法,对长春新碱耐药的SGC7901/VCR细胞系及其母细胞系SGC7901的总蛋白提取物进行测定,分析热休克蛋白27(HSP27)与MDR的关系,并对HSP27的相互作用组图进行综合研究。结果 HSP27作为一种与MDR相关的蛋白得到鉴定,并鉴定出25种HSP27相互作用蛋白。HSP27ASOs对HSP27表达的抑制可加强SGC7901/VCR对长春新碱和阿霉素的化疗敏感性。结论胃癌中HSP27与长春新碱耐受机制相关。Objective To investigate the mechanisms of multidrug resistance（MDR） in vincristine（VCR）-resistant human gastric cancer cell line SGC7901 /VCR.Methods Total protein extracts from VCR-resistant SGC7901/VCR and its parental cell line SGC7901 were tested by comparative proteomics.The association of heat shock protein 27（HSP27）,one of the highly expressed proteins in SGC7901/VCR,with MDR was analyzed using antisense oligonucleotides（ASOs） inhibition.A comprehend-sive interactome map of HSP27 was investigated by coimmunoprecipitation（IP） coupled with MS.Results HSP27 was identified as a protein showing increased expression in SGC7901/VCR.The suppression of HSP27 expression by HSP27 ASOs could enhance vincristine and adriamycin chemosensitivity in SGC7901/VCR.Identified 25 HSP27-interacting proteins by IP coupled with MS could be classified into eight categories based on their functions,which correspond to the reported functions of HSP27 with MDR.Conclusion HSP27 is relative to the mechanisms of multidrug resistance in gastric cancer.
