肝硬化大鼠肝组织生长抑素受体亚型mRNA表达的研究

摘　　要：目的　研究在大鼠肝硬化门静脉高压症形成过程中 ,肝组织生长抑素 5种受体亚型mRNA的表达变化。方法　将大鼠 30只随机分为对照组、急性坏死组和肝硬化组。提取样本肝组织总RNA ,进行逆转录 聚合酶链反应 (RT PCR)后 ,琼脂糖凝胶电泳条带经过计算机图像分析处理 ,检测大鼠生长抑素受体 (rSSTR)亚型mRNA的表达。结果　正常肝组织中rSSTR4 、rSSTR1、rSSTR5的mRNA表达量均明显高于rSSTR3、rSSTR2 (P <0 .0 5 )。急性坏死组rSSTR各受体亚型mRNA表达与对照组和肝硬化组相比均显著减少 (P <0 .0 5 )。肝硬化组rSSTR2 mRNA表达量 (16 1.97±40 .42 )较对照组 (10 5 .5 7± 37.91)显著增加 (P <0 .0 5 ) ,rSSTR1和rSSTR4 mRNA表达量 (171.33±44 .5 9、15 8.2 6± 37.79)较对照组显著减少 (P <0 .0 5 )。结论　大鼠肝硬化过程中通过激活特异的rSSTR亚型基因引起特异受体亚型mRNA的合成增加 ,从而调节rSSTRs介导的生长抑素类似物治疗肝硬化门静脉高压症的作用。Objective\ To investigate the changes in mRNA expression of subtypes of somatostatin receptors (rSSTR) during the development of portal hypertension in liver cirrhotic rats. Methods\ A total of 30 rats were randomly divided into 3 groups: control group, necrosis group and cirrhosis group. Total RNA was extracted from each sample and semi-quantitative reverse transcription and ploymerase chain reaction(RT-PCR) were used to detect the expression of mRNA of rSSTR. Results\ In the control group, all 5 subtypes of rSSTR were expressed in liver, and rSSTR4,rSSTR1 and rSSTR5 mRNA expression was significantly higher than rSSTR3 and rSSTR2 mRNA expression ( P <0.05). In the necrosis group, mRNA expression of rSSTR was significantly decreased as compared with that in the control group and cirrhosis group ( P <0.05). In the cirrhosis group, mRNA expression of rSSTR2 (161.97±40.42) was significantly increased as compared with that in the control group (105.57±37.91, P <0.05), while mRNA expression of rSSTR1(171.33±44.59) and rSSTR4(158.26±37.79) was significantly decreased as compared with that in the control group ( P <0.05). Conclusion\ The stimulated gene of specific rSSTR subtypes could increase the synthesis of mRNA of specific rSSTRs to regulate the rSSTRs-mediated effect of the analogues of somatostatin in the treatment of portal hypertension of liver cirrhosis.\;
