摘  要:目的探讨E26转录因子-1(Ets-1)和信号转导与转录激活因子6(STAT6)在食管病变组织中的表达,并分析其相关性与临床意义。方法应用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶连结法和反转录聚合酶链反应法检测36例食管鳞癌、36例癌旁非典型增生及18例正常食管黏膜组织中Ets-1、STAT6的表达。结果 Ets-1、STAT6蛋白在正常食管黏膜、癌旁非典型增生及食管鳞癌组织中阳性表达率均依次升高,组间比较差异均具有统计学意义(P<0.05),且二者呈正相关关系(r=0.447,P<0.05)。Ets-1、STAT6 mRNA在正常食管黏膜、癌旁非典型增生及食管鳞癌组织中相对含量依次升高,组间比较差异均具有统计学意义(P<0.05)。Ets-1及STAT6异常表达与食管癌的分级、浸润深度及淋巴结转移均密切相关(P<0.05)。结论 Ets-1和STAT6异常表达与食管上皮癌变有关,二者可能具有协同作用,并可作为判断患者预后的分子指标。Objective To explore expression of E26 transformation-specific-1(Ets-1) and signal transducers and activators of transcription 6(STAT6) in esophageal lesions,and to analyze the correlation with clinicopathologic characteristics.Methods The expression levels of Ets-1 and STAT6 protein and mRNA were detected in 36 cases of esophageal squamous cell carcinoma(ESCC),36 cases of atypical hyperplasia and 18 cases of normal esophageal tissues by immunohistochemistry streptavidin-peroxidase and reverse transcription-polymerase chain reaction.Results Positive expression rates of Ets-1 and STAT6 proteins were gradually increased from normal esophageal tissues,atypical hyperplasia to ESCC tissues,there were significantly difference among the groups(P0.05),and their expressions were positively correlated(r=0.447,P0.05).The relative amount of Ets-1 and STAT6 mRNAs were gradually increased from normal esophageal tissues,atypical hyperplasia to ESCC tissues,there were significantly difference among the groups(P0.05).The abnormality expression of Ets-1 and STAT6 were positively correlated with tumor grade,infiltration and lymphatic metastasis of ESCC tissue.Conclusion The abnormal expression of Ets-1 and STAT6 are related to esophageal carcinogenesis,have synergistic effect and can be used as the prognostic molecular markers.
