摘  要:目的观察卒中后抑郁(post-stroke depression,PSD)模型大鼠小脑功能的改变。方法选用健康雄性SD大鼠,随机分为4组,对照组、卒中组、单纯抑郁组、卒中后抑郁组(PSD组);并分7d、14d、21d 3个时间点进行行为学观察和测试、免疫组化法测定小脑组织功能活性依赖蛋白突触素(P38)、脑源性神经营养因子(BDNF)的表达。结果与卒中组相比,单纯抑郁组大鼠体重和糖水消耗量下降,水平运动及垂直运动水平降低(P<0.01);PSD组大鼠体重和糖水消耗量下降更为明显,水平运动及垂直运动水平降低也最为明显(P<0.01)。与卒中组比较,单纯抑郁组P38、BDNF的表达普遍减弱,与单纯抑郁组相比较,PSD组P38、BDNF的表达减弱的更为明显;组内不同时间点(7d、14d、21d)进行比较:卒中组、单纯抑郁组、PSD组随着时间点的推移,小脑组织P38、BDNF表达也呈现出减弱的趋势。结论大鼠小脑功能的改变可能与PSD的发生有关,进一步研究可能有助于阐明卒中后抑郁症的发病机制。Objective To observe the changes of cerebellar function in the post-stroke depression(PSD) model of rats.Methods The healthy SD rats were randomly divided into four groups,sham group,stroke group,depression group,and PSD group.Behavioral observation and testing were performed and the expression of P38 and BDNF in cerebellum by immunohistochemical method of on 7days,14days and 21days were detected.Results Compared with stroke group,depression rats,weight and sugar water consumption were decreased,horizontal motion and vertical motion level were lower(P〈0.01);PSD rats,weight and sugar water consumption drop were more apparent,horizontal motion and vertical motion level also reduced the most obviously(P〈0.01).Group comparison:Compared with the stroke group,in depression group and PSD group,the expression of P38 and BDNF in cerebellum were decreased.The difference between them was very significant.Intergroup comparison(7d,14d,21d):Compared with the stroke group,in depression group and PSD group on 7d,14d and 21d,P38 and BDNF were significantly differencedifferent.HE staining showed neuronal cell damage.Conclusion Cerebellar dysfunction has the potential to be involved in PSD.
