摘  要:目的研究三七总皂甙(PNS)对大鼠骨髓基质细胞骨形成蛋白-2(BMP-2)表达及碱性磷酸酶活性的影响,探讨其在骨髓基质细胞向成骨细胞增殖、分化中的作用机制。方法分离、纯化大鼠骨髓基质细胞(BMSCs),实验按不同质量浓度PNS分组(0、50、100、150、200mg.L-1),以0mg.L-1为对照组。均采用成骨诱导液培养,MTT法观察细胞增殖,ALP活性测定,Western Blotting检测细胞BMP-2表达。结果在第3、5天各组细胞增殖差异无统计学意义(P>0.05),在第7、9天,50、100、150mg.L-1组高于对照组(P<0.05);BMP-2表达水平及ALP活性在50、100及150mg.L-1组高于对照组(P<0.05),对照组少量表达BMP-2;200mg.L-1组细胞增殖、BMP-2表达水平及ALP活性与对照组差异均无统计学意义(P>0.05)。结论三七总皂甙可通过上调BMP-2的表达水平,增强碱性磷酸酶活性,促进骨髓基质细胞向成骨细胞增殖、分化。Objective To study the effect of total saponins of panax notoginseng (PNS) on the expression of bone morphogenetic protein-2 (BMP-2) and alkaline phosphates (ALP) activity in rat bone marrow stromal cells (BMSCs), and to elucidate the mechanism of PNS action on the proliferation of BMSCs during differentiation into osteoblasts. Methods After isolation and puri- fication,rat BMSCs were induced in medium alone (control) or osteogenic medium containing dif- ferent concentrations of PNS (50,100,150 and 200 mg · L-1 ). The proliferation of BMSCs was determined by MTT assay. The osteogenic differentiation was evaluated by ALP activity. BMP-2 expression was detected by Western blotting. Results There were no significant differences in theproliferation of BMSCs at 3 and 5 days after treatment among the different groups (P〉0.05). Compared with control group, the proliferation of BMSCs, BMP-2 expression and ALP activity were significantly increased by treatment with 50,100 or 150 mg · L-1PNS at 7 and 9 days after treatment (P〈0.05). However,no significant differences were found in cell proliferation,BMP-2 expression and ALP activity between 200 mg · L-1 PNS treatment group and control group (P〉0.05). Conclusion The PNS can promote the differentiation of BMSCs into osteoblasts through increasing BMP-2 expression and ALP activity.
