肝细胞癌癌组织α-1,3岩藻糖基转移酶各亚型mRNA的半定量研究

摘　　要：目的　探讨肝癌组织α 1,3岩藻糖基转移酶 (α 1,3 fucosyltransferase,FucT)的各亚型Ⅲ、Ⅳ、Ⅴ、Ⅵ、Ⅶ的mRNA表达及其与肝癌转移的关系。方法　应用异硫氰酸胍一步法获得无DNA污染的RNA ,经RT PCR扩增后对各亚型FucTPCR产物测序鉴定并扫描定量。结果　肝癌组织未发现FucT Ⅲ、ⅤmRNA的表达 ,FucT Ⅶ在癌和癌周组织的表达都很低 ,且两者无显著性差异。肝癌组织FucT Ⅳ、Ⅵ的表达明显高于癌旁肝组织与正常肝组织 ,转移性肝癌FucT Ⅳ、Ⅵ的表达显著高于非转移性肝癌者。结论　FucT Ⅳ、Ⅵ与肝癌的转移有关。Purpose To study the mRNA expression of different subtypes of α?1,3 fucosyltransferase (FucT?Ⅲ,Ⅳ,Ⅴ,Ⅵand Ⅶ)and reveal its relation to the metastasis of hepatocellular carcinoma. Methods Purified RNA was used in RT?PCR,the identification of the PCR products by DNA sequencing and the semiquantitative analysis was performed by VDS image systen. Results FucT?Ⅲ,Ⅴsubtypes mRNA were not detected in this series.FucT?Ⅶ mRNA expression was low and there was no significance difference bet ween tumor and surrounding tissue.The expression of FucT?Ⅳ and Ⅵ were higher in tumor than in surrounding tissue and normal liver.These two FucT subtypes also increased higher in the metastasis group than in the group without metastasis. Conclusions FucT?Ⅳ and Ⅵ might be related to the metastasis of hepatocellular carcinoma.
