预防性给予重组人红细胞生成素对神经病理性疼痛大鼠的行为学影响

摘  要:目的神经病理性疼痛难于治疗,探寻新的治疗药物是未来神经病理性疼痛治疗的重要研究方向。文中探讨雄性大鼠L5脊神经切断前预防性给予重组人红细胞生成素(Recombinant Human Erythropoietin,rhEPO)对大鼠机械、热痛觉高敏的影响及可能机制。方法选用L5脊神经切断神经病理性模型,将50只SD雄性大鼠随机分成5组(每组10只),分别为:假手术组、损伤腹腔注射等渗盐水组(等渗盐水组)、损伤腹腔注射rhEPO 1000 U/kg组(rhEPO 1000 U/kg组)、损伤腹腔注射rhEPO 3000 U/kg组(rhEPO 3000 U/kg组)、损伤腹腔注射rhEPO 5000 U/kg组(rhEPO 5000 U/kg组),术前1 d给药,连续给药7 d。采用von Frey和Hargreaves法测定各组大鼠机械缩足反射阈(mechnical withdrawal threstholds,MWT)值和热缩足潜伏期(thermal withdrawal latencies,TWL)。结果与等渗盐水组大鼠相比,预防性腹腔注射rhEPO 3000 U/kg或5000 U/kg,能明显缓解大鼠的机械、热痛高敏行为。结论预防性给予rhEPO(3000 U/kg或5000 U/kg)能预防神经病理性疼痛的发生。Objective Neuropathic pain is a refractory disease, and searching for new therapeutic drugs for it remains an im- portant task for future studies of the disease. This article is to investigate the effect of intraperitoneal administration of recombinant hu- man erythropoietin (rhEPO) before L5 spinal nerve transection (SNT) on mechanical and thermal hyperalgesia in rats. Methods We made rat models of neuropathic pain by L5 SNT and equally randomized 50 male SD rats to a sham-operation group, an Ls SNT + saline group given isotonic saline, and 3 L5 SNT + rhEPO groups treated with rhEPO at 1000, 3000 and 5000 U/kg, respectively, ad- ministered intraperitoneally 1 day before surgery for consecutive 7 days. We measured the hind paw mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) of all the animals at 7 days after surgery using von Frey hairs and Hargreaves tests, respectively. R^ults Intraperitoneal administration of rhEPO at 3000 and 5000 U/kg significantly attenuated mechanical and ther- mal hyperalgesia of the model rats as compared with the saline controls. Conchmion Administration of rhEPO at 3000 or 5000 U/kg can prevent neuropathic pain induced by Ls spinal nerve transection.
