人源抗胃癌单链抗体的构建及初步鉴定

摘  要:目的构建库容大、多样性好的噬菌体展示人源抗胃癌单链抗体。方法收集胃癌患者外周血分离淋巴细胞,利用噬菌体展示技术,构建人源单链抗体库并初步筛选抗胃癌单链抗体,用ELISA方法检验该抗体与胃癌细胞结合活性。结果成功构建了次级库容达4.0×109pfu/ml的噬菌体展示人源单链抗体库,并初步筛选到与胃癌细胞特异性结合的单链抗体。结论成功构建和筛选人源抗胃癌单链抗体,为进一步表达和应用特异性抗胃癌单链抗体奠定基础。Objective To construct high - capacity phage display library of human single - chain variable fragment (scFv) against gastric carcinoma. Methods Lymphocytes were selected from patients of gastric carcinoma and separated by density gradient centrifugation. Then the scFv gene library was constructed by phage display technology. Results We successfully constructed human phage - displayed scFv library with a titer of 4. 0 × 109 pfu/ml. And ELISA verified that the scFv against gastric carcinoma could specifically conjugate with gastric carcinoma cell. Conclusion High - capacity phage display library of human scFv was constructed successfully. It shed light on the expression and application of the specific scFv antibody against gastric carcinoma in the following study.
